Objective:To explore the prevention and cure function among jianpiyifeifang, huatanfang, jianpiyifeihuatanfang, and the impact of underlying problem, the symptom or both the symptoms and root causes by establishing the rat model with COPD. Im around goblet cells hyperplasia, high airway mucus hyper, airway inflammation, such as signal transduction, from EGFR, TNF-α, NF-κB, PI3K/AKT signaling pathway to discuss on the spleen lung phlegm party COPD model rats, the differences of treatment based on the mechanism of action and sputum specimens.Method:The model rats of COPD were established by pure given cigarettes fume atmosphere. After that, all the subjects were randomly divided into six groups including blank group, model group, jianpiyifei group, huatan group, jianpiyifeihuatan group and Carbocisteine group. Jianpiyifeihuatan and its separated parts were chosen as the administration drugs and the blank group was administration with pure water as well as model group.1. The pulmonary function tests was taken to test six groups rats’ pulmonary function; With arterial blood treated of anticoagulation, PaCO2, PaO2, PH and SaO2 and other arterial blood gas indicator in rats were measured by arterial blood gas analysis.2. ELISA method was separated to test the content of mucins of MUC5B、NE and ROS in the bronchoalveolar lavage fluid of rats.3. Western Blot method was separated to measure the phosphorylation level of EGFR、PI3K and AKT in the lung tissues of rats.4. With the internal reference for the gene expression of control, CLCA1, EGFR, NF-κ BmRNA in the lung tissue were observed by PCR.Result:1. Lung function and Aterial blood gas analysis:compared with the blank group, the relevant indicators were abnormal conditions in model group:the level of FEV0.3/FVC, pH, PaO2, SaO2 were decreased, the PaCO2 was increased; compared with the model group, the above relevant indicators had been improved including Jianpiyifei group, huatan group, jianpiyifeihuatan group, especially the Jianpiyifeihuatan group.2. The result of ELISA:compared with the blank group, the content of mucins of MUC5B、 NE and ROS in the bronchoalveolar lavage fluid of rats in model group were abnormal increased, and there were significant difference between the two groups. The result showed that goblet cells hyperplasia and airway mucus hyper were enhanced in rats of COPD. Compared with the model group, the content of mucins of MUC5B、 NE and ROS in the bronchoalveolar lavage fluid of rats in Jianpiyifei group, huatan group, jianpiyifeihuatan group were decreased. It showed that a series of jianpiyifeihuatanfang can restrain from signal transduction pathway of goblet cells hyperplasia and airway mucus hyper in rats of COPD.3. The result of Western Blot:Compared with the normal group, the expressions of phospho-EGFR, phospho-PI3K, phospho-AKT and phospho-38 in model group rats were higher. Compared with model group, the expressions of phospho-ECFR, phospho-PI3K, phospho-AKT and phospho-38 in all treating group rats were lower.4. The result of PCR:Compared with the normal group, the expressions of CLCA1mRNA、 EGFRmRNA and NF-κBmRNA in model group rats were higher. Compared with model group, the expressions of CLCA1mRNA、EGFRmRNA and NF-κ BmRNA in all treating group rats were lower, but it also higher than the normal group.Conclusion:In summary, Jianpiyifeihuatanfang and its separated parts could decrease the content of inflammatory cytokines, so as to suppress the goblet cells metaplasia and mucus hypersecretion, to improve the lung function of model rats of COPD through inhibition of MAPK/TNF-α/NF-κB signaling pathway. But there are some differences among three Traditional Chinese Medicine group:the best is Jianpiyifeihuatan group, then group is Jianpiyifei group, last group is the huatan group.
