Study On The Expression Of Related Molecules In Allergic Treg Cells And The Intervention Mechanism Of Allergic

Objective:Looking for the effective specific molecular markers of the Allergic Constitution by comparing the associated moleculars of Treg cell differentiation between the Allergic Constitution and the Balanced Constitution. To find out the biological target of treating allergic diseases with the experienced formula "GuoMinKang" by interfering the Allergic Constitution. From the perspective of TCM constitution theory, to explore the way to treat allergic diseases, expecting to give an further guidance for clinical treatment.Methods:Basing on the Classification and Determination of Constitution in TCM and the allergen test, we recruited 44 subjects who were physical examinees from TCM constitution center of Beijing University of Traditinal Chinese Medicine Guoyitang Outpatient Department etc and between 10 to 40 years old. Among the subjects,24 ones belong to the Allergic Constitution as experimental group,which role out other diseases diagnosed by modern medicine,women who were pregnant or in lactation,and 20 in Balanced Constitution as control group. Ensure that the baseline data of each group were comparable,and make sure the experimental group won’t take other anti-allergic drugs.Signed the informed consent forms before intervention. The Balanced Constitution subjects and the Allergic Constitution ones before and after intervention were draw blood respectively,collect 10 ml blood early in the morning on an empty stomach condition,36ul whole blood was pretreat by QuantiGene place kit instructions, the rest of the blood was separated out serum and saved in-80 degrees freezer. The serum was for detecting protein expression levels of IL-6,IL-10,TGF-β,IL-2 by protein chip; pretreated whole blood for testing gene expression levels of IL-10mRNA, IL-6RmRNA, IL-2RBmRNA, ITK mRNA, IL-4R mRNA by QuantiGene Plex. Data analysis using SPSS 17.0 software processing, differences between the Allergic Constitution and the Balanced Constitution with One-Way ANOVA,differences between the Allergic Constitution before and after intervention with paired sample t-test analysis, the data were expressed as meanistandard deviation (x±s), P< 0.05 says statistically significant, P< 0.01 means a significant statistical difference.Results: ①The Allergic Constitution before intervention,compared with the Balanced Constitution Volunteers, we found IL-6R、IL-2 protein expression levels increased and there are significant differences (P<0.01);IL-10、TGF-β protein expression levels elevated but not statistically significant (P> 0.05).It found that the expression levels of IL-4RmRNAs、ITKmRNA IL-2RBmRNA,three genes all are higher than the Balanced Constitution Volunteers but make no statistically significant change(P>0.05). And the expression levels of IL-10mRNA、IL-6RmRNA, both two genes are lower than the Balanced Constitution Volunteers but make no statistically significant change(P>0.05)②The Allergic Constitution after intervention, compared with that before intervention,we found IL-6R protein levels gained and there was a significant difference (P<0.01); IL-2、IL-10、TGFβ，those three protein levels all decreased and there was no significant difference (P> 0.05).It found that IL-6R mRNA、IL2RBmRNA、ITKmRNA expression levels reduced and IL-10mRNA、 IL-4R mRNA all elevated but those were not statistically significant (P> 0.05).Conclusions: ①IL-6R、IL-2 are significant different factors between the Allergic Constitution and the Balanced Constitution ② The pathway of "GuoMinkang" to treat the Allergic Constitution may be associated with IL-6R,more specific mechanism needs further exploration.③The most meaningless experimental datas may be related to Treg cell plasticity,protein-protein interactions,the specificility of The AllergicConstitutions and so on.
