Purpose:With the method of serum pharmacology, to the effect of angiotensin II as a stimulating factor to observe Jiawei Yushen Decoction Pharmacological Serum on cultured rat glomerular mesangial cell proliferation and correlation factors.To investigate the effect of Jiawei Yushen Decoction on mesangial proliferative glomerulonephritis. Material and method: 1.Select the SPF level of 30 healthy SD rats, half male and half female.The rats were randomly divided into blank group, western medicine group, Jiawei Yushen Decoction low, medium and high dose group. After 2 days of adaptive feeding, the traditional Chinese medicine group were given the corresponding traditional Chinese medicine, the western medicine group was treated with valsartan, and the normal group received the same amount of normal saline for 5 days. 1 hours after the last administration, blood was collected from abdominal aorta, and the pharmacological serum was prepared. 2.Rat glomerular mesangial cell recovery and passage, collected cells.Cells were divided into blank control group, western medicine group, model group, Jiawei Yushen Decoction low, medium and high dose group shop in 96 well plates. With Ang II as a model stimulating factor, each group added serum,trained in incubator at 37℃, 5%CO2. After 48 hours the cells proliferation was measured by MTT assay. The levels of IL-6 and MCP-1 in glomerular mesangial cell supernatant were detected by enzyme-linked immunosorbent assay(ELISA). Results: 1.The effect of Jiaweiyushen decoction on proliferation of mesangial cells of rats induced by AngⅡResults show that,compared with the blank group,the number of modle group was incresed had a statistical difference(p<0.01).It indicated that Ang can promote rat Ⅱglomerular mesangial cell proliferation.Compared with model group,the mesangial cells number of jiaweiyushen decoction high, medium,low dose group is reduced. The difference was statistically significance(p<0.01),suggesting that Jiaweiyushen decoction and valsartan can inhibit rat glomerular mesangial cell proliferation induced by AngⅡ. 2.The effect of Jiaweiyushen decoction on rat glomerular mesangial cells secrete IL-6The blank group IL-6 in low concentration, there are significant differences compared with the model group(p<0.01).The concentration of model group was 2 times that of the blank group. Compared with the model group, the concentration of IL-6 in the high, medium,low dose group and Western medicine group was significantly lower than that in the model group,and the differences was considered statistically significant(P<0.01).At the same time, there is a dose-response relationship between Jiawei Yushen Decoction high, medium and low dose group. 3.The effect of Jiaweiyushen decoction on rat glomerular mesangial cells secrete MCP-1The blank group MCP-1 in low concentration, there are significant differences compared with the model group(p<0.01).The concentration of model group was 2 times that of the blank group. Compared with the model group, the concentration of MCP-1 in the high, medium,low dose group and Western medicine group was significantly lower than that in the model group,and the differences was considered statistically significant(P<0.01).At the same time, there is a dose-response relationship between Jiawei Yushen Decoction high, medium and low dose group. Conclusion: 1.Jiwei YuShen decoction can inhibit the proliferation of rat glomerular mesangial cells induced by AngⅡ. 2.Jiawei Yushen decoction can inhibit the IL-6 and MCP-1 excessive secretion in rat glomerular mesangial cell supernatants induced by Ang II. 3.By inhibiting the proliferation of glomerular mesangial cells and excessive expression of inflammatory cytokines in rats, the effect mechanism of the treatment of mesangial proliferative glomerulonephritis was demonstrated.
