Objective:Cardiovascular and cerebrovascular diseases have become the leading cause of death, at least seven million Chinese people have stroke. Now, stroke has causing significant health burden.High homocysteine hypertension accounted for about 75% patients with high blood pressure in China. Recently about the relationship between Hey and intracranial hemorrhage(ICH) articles is controversial. Therefore, we investigate the relationship between Hey with intracranial hemorrhage stoke in hypertensive mice.Methods:Eight-month-old male C57BL/6 mice administered angiotensinâ…¡ (Angâ…¡) to produce hypertension and fed with L-NAME and homocysteine(Hcy) for 18 weeks to induce hemorrhage stroke.80 mice were divided into four groups:Hey+Angâ…¡; Hey+ Angâ…¡+L-NAME; Angâ…¡+L-NAME; Angâ…¡. Angâ…¡ used to raise blood pressure in mice, L-NAME was arginine synthase inhibitors for inducing cerebral hemorrhage. The first to use water containing Hey in mice fed six weeks. At seven weeks, embedding Angâ…¡ in mice back subcutaneous and at the same time treatment with L-NAME (100 mg/kg per day; Sigma-Aldrich) in drinking water. If behavior phenomena of cerebral apoplexy ocured, the mouse was in was anesthesiaed, preserved in the brain and serum. The feeding time was 18 weeks. Describe the relationship between Hey and intracerebral hemorrhage stroke by Kaplan-Meier plot curve. BP-2000 measuring blood pressure in mice; Automatic biochemical analyzer measurement Hey concentration in blood in mice; MR monitoring bleeding site location in mice; Measuring the mice bleeding area by HE staining; Immunohistochemical staining is used to calculate bleeding site of vascular smooth muscle cell number.Result:Mice fed Hey six weeks later, Hey blood levels rise to 23.07 umol/L, after 18 weeks average Hey concentration in blood of mice was 41.51 umol/L; Blood pressure between 170-190. The number of each group of mice die in cerebral hemorrhage:Hey+ Angâ…¡ group 2 only; Hey+Angâ…¡+L-NAME group 10; Angâ…¡+L-NAME group 10; Angâ…¡ group 0. Hcy+Angâ…¡+L-NAME and Angâ…¡+L-NAME group survival curve of the p value is 0.162.Conclusions:The result shown that hyperhomocysteinemia may be has no effect on the occurrence of hypertension intracerebral hemorrhage in mice.Objective:H-type Hypertension accounts for about 75% of adult patients with hypertension, high levels of plasma homocysteine (Hcy) is an independent risk factor for cardiovascular disease. Existing studies have shown that by taking folic acid to reduce homocysteine levels could reduce the incidence of stroke, but there were some controversial. Therefore, this study aimed to investigate folic acid and hemorrhagic stroke on animal level.Methods:Eight months C57BL/6 mice were divided into H-hypertensive cerebral hemorrhage group(20) and H-hypertensive intracerebral hemorrhage+folic acid group(17). When stroke behavior occurs, the mice were sacrificed and brain tissue preserved for HE staining and immunohistochemical staining to determine the mice bleeding site. After 18 weeks, all mice were sacrificed. Use SPSS 19.0 analysis of K-M curve mice relativities between the two groups.Results:Through behavior and the final pathology testing found that H-type hypertensive group 10 died of hypertension cerebral hemorrhage; H-type hypertension+ folic group 13 died of a brain hemorrhage. After comparing the survival curves of the H-hypertension group and H-hypertension+folate group found no difference between the two groups, p=0.127.Conclusion:Folic acid cannot reduce the risk of hemorrhagic stroke in H-hypertension mice.Objective:Hypertensive intracerebral hemorrhage is a common type of stroke, although its incidence was lower than those of ischemic stroke, but its onset disaster, prognosis is poorer. At present, cerebrovascular pathological changes after hemorrhage are poorly understood. This study proposed use hypertension cerebral hemorrhage mouse model to study hypertension cerebral hemorrhage mechanism of vascular lesions.Methods:Eight months male C57BL/6 mice were divided into two groups, one group of hypertensive intracerebral hemorrhage group, the other group was the control group. When stroke behavior occurs, the mice were sacrificed and brain tissue preserved. HE staining to determine the bleeding site, using elastic fibers staining observe of vascular structures of bleeding site.Results:Elastic fiber dyeing found a ruptured blood vessel hemorrhage parts, straightening, tension disappearance of internal elastic membrane. We also found blood vessels burst to internal, causing false lumen.Conclusion:This study found that the change of elastic lining may be associated with the occurrence of hypertension cerebral hemorrhage in mice.
