Objective:Metabolic syndrome prescription is a clinical prescription for treatment of metabolic syndrome. It contains astragagalus, trichosanthis and other herb medicines.it controls blood sugar, blood lipid metabolism and blood pressure.This experiment optimizes the water extraction technology of metabolic syndrome prescription to make it into granules.Method:1. Quality control of original herb medicineMeasure the content of all the herb medicine in prescription respectively by the method of pharmacopoeia to ensure the quality of materials.2. Optimization of preparation technologyWater extraction part:In order to select the best extraction process of metabolic syndrome prescription, we design orthogonal test to explore the extraction duration, solvent dosage and times of extraction based on the content of astragalus glycosides,calycosin glucoside,linarin and hyperoside and verify the process after the selection.Then we screen the concentration of ethanol and relative density of the inspection to optimize the optimal ethanol sinking process.Ethanol extraction part:In order to select the best extraction process of metabolic syndrome prescription, we design orthogonal test to explore the ethanol concentration, solvent dosage and times of extraction process based on the content of specnuezhenide and alisol B 23-acetate and verify the process after the selection.3. Pharmacological experimentAscertain the best extraction process through pharmacological experiments.4. Research on granulating technologyExplore the granulating technology of metabolic syndrome prescription through the amount of excipient, solvent dosage and ethanol concentration.5.Research on the quality standards of the metabolic syndrome prescriptionIdentify all kinds of herb medicine in prescription respectively by TLC identification according to pharmacopoeia.Using HPLC and UV-detecortor to determine the content of privet glycosides and hyperoside.Results:1.preparation technology:The best water extraction process:use 12 times water of all the herb weight to extract for three times which last 2.5 hours. Combine water extraction from each time and concentrate the solution to relative density of 1.068-1.080. Add 95% ethanol to the extraction and dilute until the ethanol concentration of the solution reach 50%, place for 24 hours, filtrate it.The best ethanol extraction process:use 6 times 80% ethanol of all the herb weight to extract for 3 times which last 2 hours.Concentrate the solution,filtrate.Desiccate two parts of the filtrate under reduced pressure in 60℃.2.Pharmacological experimentCompare three extraction processes.
