“Taisui”is unidentified biological object which is not clearly defined and classified up to now. Its existence, function and influence have always been controversial. The research results in scientific community fail to explain the biological evolutionary status of Taisui. This study discusses the classification status of Taisui from both macroscopic and microcosmic aspects with Taisui LNUT 008 as the raw material. The result is as the following:The apparent biological characters of“Taisu”i are stated briefly:“Taisui”feels like meat and does not have chlorophyll. Tissu of“Taisui”cannot be broken into pieces after drying out and can be melted in high temperature water bath. Besides, it has high moisture content. By using scanning electron microscope and transmission electron microscopy, there came the experimental result. It indicates that Taisui LNUT 008 contains spherical and lumpish regular crystalline substance outside surface and polycystic cellular double membrane structure inside. But the typical eukaryotic cell structure and mycelium were not observed.The attempts making use of animal, fungi and bacteria molecular biology kit and the other extraction methods with different lysis buffer to extract genome DNA of Taisui did not gain effective DNA strip.This study has sought and optimized the genome extraction method. It has gained the“Taisui”genome DNA through the innovation of a kind manual extraction method of genomic DNA. The experiment has conducted an identification of 16S rDNA, 18S rDNA and ITS using“Taisui”genome DNA as formwork in PCR amplification system. The amplification result indicates there is no amplification aim strip.In order to explicit the classification status of Taisui, the experiment constructs the Taisui genome library starting on Taisui genome DNA and gets part DNA order of Taisui to make comparative analysis identification. The fragment which is got through cutting of“Taisui”genome DNA by Sau3AI random enzyme connects the pUC18 plasmid vector that got through BamHI and dephosphorylation and then they are transferred into the E.coliDH5αhost cell to construct part“Taisui”genomic library. Plasmid is identified and the different length of insert fragments can be gained. Sequencing is conducted on the effective insert fragments and then BLAST comparative study is carried with the nucleotide sequence in GeneBank database. The result indicates there is no homology of the fragments and the fragment in data base. From the above experiment result, it is can be seen there is possibility Taisui become a new kind of undefined species in the evolution history.This study makes dramatic breakthroughs in the aspect of the biological character and systematic position of“Taisui”; at the same time, it lays an important foundation for the reveal of Taisui’s essence and signifies much for the research of Taisui in future.
