Energy is the most important problem in21th century, as contain of fossil fuel is reducing day by day, and the serious environment consequences followed improper using, making people be embarrassed in chasing for economy development and protecting environment, according to these sciences are trying hard to solve two main problems:one is how to, the other is to find out kind of new energy to feed our need. As a researcher in biology, this article was established in Biological field, researched biological hydrogen energy, taken Chlamydomonas reinhardtii wild type CC125as mold species, and studied the mechanism of releasing hydrogen by steps, then found out the possible resource of proton and proved by experiments.By analyzing the two possible ways of proton origins:one is acetate, the other is water, we devised three main parts to test and verify. Firstly, ion chromatograph was used to monitor formic acid and acetic acid in medium during its growing phase and sulfur deprivation phase and to watch the way they change, then time by time the water in medium was taken up by deuteroxide, and finally no water in cells. Although it was hard to achieve, we got some meaningful data then designed the third part of experiment, which isotope labeled acetate was applied to culture Chlamydomonas reinhardtii, at last tested the hydrogen to define the source. For this is a kind of exploratory research, in this limited time, study can only get to the second part, and need further research by successors. At present we can figure out:as for first part, during the growth phase, formic acid can apparent, and acetate declined markedly, while in sulfur deprivation phase, formic acid increased steadily, and acetate descended but remained some content. Using SPSS we gained some useful correlativity. To the second part, Chlamydomonas reinhardtii was stimulated with deuteroxide, found out that deuteroxide can restrict the growth of cells, and this showed much obviously when rate goes to60%. Analysing the results we discover40%will make the most activity of PS Ⅱ, maybe contain highest chlorophyll at50%, and the shape of cells have changed greatly, so as its motor ability, yet the mechanism leads to these needs to further research.
