The core fucosylation modification of the N-glycosylation protein is known tobe involved in many important biological functions.Recent studies on N-glycoproteinvariants of the tumor cell surface have revealed that this phenomenon happened withabnormal fucosylation,and many core fucosylation is now already serves as tumormarkers.such as the increase of fucosylated AFP is accompanied by an increase infucosyltransferases’expression.The relationship bewteen the abnormal and the development of the tumor havecaused the glycobiologists’attention.But the lack of the fucose probe moleculeslimited the research of the biological functions.This essay was designed to thesynthesis of fucosylated N-Glycans probes,including the fluorophore-labeled probesand the Glyco-agarose beads combined probes.The fluorophore-labeled probes can beused to the selection of the fucose-binding lectin in vivo or in vitro,and theGlyco-agarose beads combined probes can be used for the extraction,separation andidentification of biological lectin.We have finished the synthesis of the fucosylated trisaccharride GlcNAc-GlcNAc(Fuc) and the synthesis of the disaccharide GlcNAc-GlcNAc. Currently, wealso used the click chemistry methods to introduce the disaccharride to thefluorophore,and we have already got the fluorophore-labeled probes.
