Massive green tides have been occurring six years in the coast of theYellow Sea, China. The green tide expanded to58000km2(floating area)and the calculated covering area was about2100km2in2009. It has beenaffecting the marine ecological environment seriously. Utilization of greentides become a research focus. Ulva prolifera is the dominant species ofgreen tide, and using Ulva prolifera as cosmetic ingredients is a newdirection. In this paper, we study the effect of polysaccharides from Ulvaprolifera on anti-ultraviolet irradiation, antioxidant, moisture absorptionand retention, for potential application on cosmetic ingredients.The crude polysaccharides were obtained from Ulva proliferacultivated in Jiangsu using water extraction and ethanol precipitation. Thepure polysaccharides were purified by DEAE-52column and SephadexG-25.1、To study the protection of polysaccharides on human skin fibroblasts(HSF) cell, HSF cell was exposed to H2O2in vitro to establish oxidativeinjury model, cell ROS, MDA, CAT, GSH-PX were measured. We chose2m mol/L as the final concentration of H2O2. When the concentration ofpolysaccharides was0.5mg/mL, the rate of the proliferation reached154.4%. The pure polysaccharides (UPP1-1, UPP1-2, UPP2-1and UPP2-2)have a better protective effect on human skin fibroblasts cell than0.5mg/ml crude polysaccharides, among which UPP1-1is the best, with theproliferation rate achieving174.6%, compared with the antioxidantindicators (MDA, CAT, GSH-PX, cell ROS).It was indicated that thepolysaccharides have the capacity to protect human skin fibroblast from H2O2damage and enhance the free radical scavenging ability of HSF cell.2、The anti-ultraviolet irradiation, antioxidant, moisture absorption andretention were measured. Firstly, crude polysaccharides have the capacityto protect human skin fibroblast cell from UVB damage, beatingMentholatum sunscreen spray (Sun Protection Factor30+) onanti-ultraviolet irradiation. Secondly, crude polysaccharides scavengedDPPH by92.11%at1.5mg/mL and hydroxyl radical by83.40%at3mg/mL. Finally, at humidity level of55%and85%, crude polysaccharideswere demonstrably superior to glycerol and Qianxiancao loofah water onmoisture absorption. Furthermore, on moisture retention, a mixture ofcrude polysaccharides and glycerol (1:1) was superior to others at humiditylevel of43%and55%.The results implied that polysaccharides from Ulva prolifera have thecapacity to protect human skin fibroblast from H2O2and UVB induceddamage. It was indicated that crude polysaccharides play an important rolein anti-ultraviolet irradiation, antioxidant, moisture absorption and retention.Therefore, the polysaccharides from Ulva prolifera have the potential ascosmetic ingredients.
