Collagen is an abundant protein in vertebrates and fishes, they have been widelyused in medicine, chemical industry, etc.Extracting collagen from waste production andstudying their performance can not only reduce environmental pollution, increase theadded value of industrial production, but also provide theoretical basis for theapplication of collagen. For example, people more and more demand for pearls with thedevelopment of economy, to further promote the development of the pearl breeding. Butwhen the pearl was taked away from the pearl mussel, the rest were discarded or wereused as feed, which caused the environmental pollution and resource waste directly. Asthe pearl mussel contains large amounts of protein in the body, if the collagen can beextracted from the pearl mussel not only increase the added value of pearl mussel, butalso reduce environmental pollution.In this paper, collagens were extracted via citricacid, acetic acid-pepsin and acetic acid-pepsin-ultrasonic process from the pearl mussel.The extracted collagens were studied by production comparison, SDS-PAGE, ultravioletspectrum, FTIR, Zeta, viscosity and solubility. In addition, the extracted collagen withacetic acid-pepsin were studied by modifying, providing the theoretical basis forextraction and actual application.1. According to the principle of acid soluble collagen, using0.5M citric acidextracted collagen. It was found that the extracted collagen has high purity, high yield,and keep a complete triple helical structure, extraction process is simple and rawmaterials is cheap.2. Based on enzyme catalysis and digestion, and acid soluble principle, using0.5Macetic acid solution and pepsin extracted the enzyme-solubilized collagen, it was foundthat the yield of collagen had a very big enhancement compared with acid-solubilizedcollagen. The collagen keep the triple helical structure. The nature of collagen wassimilar toⅠtype collagen, which suggested the existence of pepsin has a certain role inpromoting to extraction of collagen, but time increased the cost of extraction.3. According to the ultrasonic vibration and thermal effect, on the basis ofenzyme-solubilized collagen joined the ultrasound. Extracting the collagen according todifferent vibration and thermal effect of different ultrasonic, and then study them. It wasfound that: under the suitable ultrasonic power, join the ultrasound can not only significantly increase the production of collagen, and can maintain the completestructure of collagen, but also can improve the solubility of collagen. Howere, when theultrasonic power is too low, the ultrasonic has not the role of acceleration, it will cause awaste of resources, and when the ultrasonic power is too high, the cavity effect andthermal effect of ultrasound can make collagen denaturation, structure changes and theyield decreased.4. According to the nature of the enzyme-solubilize collagen, a simple modificationof the collagen was made. Add appropriate zirconium silicate in the collagen solutioncan make the viscosity and denaturation temperature of collagen has a greatlyimprovement, provide a broader prospect for the application of collagen.
