Telomerase in human immortalized cell field has always been a hot spot,Discussion has been immortalized cell proliferation of telomerase abnormalities for the diagnosis and treatment of many diseases provide new ideas.Telomerase as life "biological clock" in the course of embryonic development has played a crucial role.While maintaining chromosomal stability and cell viability and can extend the shortened telomeres.Therefore, telomerase for somatic cell of cloned embryo into research has important significance.In this experiment, to be used293positive cells as standard positive control,cloned embryos of mice as experimental material.Compare the natural mating early embryo to cloned embryos of telomerase activity by TRAP-gentation.The experimental results are as follows:1.Positivecells in the controlstandardstrips,Telomerase activitywas positively correlatedwith theconcentration of cells.2.Mouse oocytes,In vivofertilized embryos and Eachcellof thereconstructed embryos were detected aspositive fortelomerase.Telomerase activitywithembryosplittingupward trend,The highestinthe blastocyst stage.Zygote(1-cell)of relatively high telomerase activity.3.Comparison ofreconstructed embryosfertilized embryosin vivo,2-cell telomerase activity relative differences(P<0.05),no significant differences in4-cell,8-cell,morula,blastocyst stage(P>0.05).4.The same sampleof2-Cell4-Cell8-telomerase activitywas not significant(P>0.05).Blastocyst stageof embryonictelomerase activitywithsignificant differences(P<0.05).5.Single blastomeres overall downward trend in the relative activity was to reach the lowest at blastocyst.embryo Single-cell during the2-cell significant differences with other Period(P<0.05).6.Through the in vivo fertilized embryos and blastocysts somatic cell count during the reconstructed embryos, in vivo fertilized embryos than single blastomeres somatic cells of the blastocyst reconstructed embryos, and there was a significant difference (P<0.05).
