:To analysis of GPI-PLD gene and its expression product proteins by bioinformatics technology and other different biological methods, analysis of systems of the different characteristics of these proteins, the focus of analysis of the human GPI-PLD gene and its enzyme protein structure.Search for gene and protein databases, such as IntEnz, Swiss-Prot ExPASy databases. Combined retrieve relevant papers such as Medline, PROSITE Database. After obtained the data, the use of networks and the software, such as study of gene and protein-related software, MEGA5, ClustalX, PAUP, PHYLIP. the structural characteristics of the different biological GPI-PLD gene sequence, cDNA sequence and expression of the gene, its enzyme protein sequence, as well as the structural characteristics of the enzyme protein homologous and evolutionary trees was analysised.1. There are more than30species GPI-PLD gene in vertebrates, and the orangutans of the gene located in chromosome6for human, GPI-PLD gene of the human genome is70791bp in length, including25exons,24introns. Bacteria do not have this gene.2. Analysis18kinds of (focus on analysis of seven kinds) biological GPI-PLD DNA sequence, the highest homology between man and chimpanzee was found, and the lowest homology found chicken. GPI-PLD gene nucleotide variation to single nucleotide polymorphisms (SNPs) are the most common. The type of variation is mainly related to synonymous mutations and missense mutations.3. There are more than20kinds of biological expression of GPI-PLD mRNA or enzyme protein. Organs of expression related to bone marrow, pancreas and other organs, as well as RAW-264.7, SW-742cell line. There are GPI-PLD of expression in subcellular organelles including the plasma membrane, Golgi apparatus, and secretory vesicles. Human GPI-PLD cDNA full length is of more than2800bp. The human bone marrow, pancreas and liver cDNA clones have homology of more than95%.4. Different organisms according to their putative gene or experiment confirmed the GPI-PLD enzyme molecules are in a polypeptide chain, the length of the different organisms is same or differents, homologous Phylogenetic analysis with the human showed that, as the different organisms, the walrus evolution is more nearly, the white Whiskers gibbon volutionary is farthest.5. GPI-PLD enzyme molecule is a glycoprotein may also be phosphorylation. GPI-PLD has7repeat peptides, the three calcium binding sites,8glycosylation sites, one zinc-binding site present in the enzyme molecule.6. Secreted GPI-PLD in the determination of its enzymatic activity must be added to the ionic detergents, the optimum pH is for the pH5-pH8. Suramin, ionic detergent, etc., is an activator of the enzyme, and the phenanthroline, lysophosphatidic acid is an inhibitor of the enzyme.Retrieving genes, proteins, and related papers database, access to the large number of GPI-PLD information, analysis or software to analyze the presence and expression of the gene of species, the structural characteristics of the human gene expression obtained after homologous with other organisms and so many interesting findings.
