Velarifictorus micado(Saussure),Nickname “ququ”, belonging to Insecta, Orthoptera, Gryllidea, Velarifictorus,one of the most common cricket varietie used to fighting. There are a lot of cricket origins in china,but ITS different among geographic populations.The two biggest origins are Dezhou NingJin County and Taian Ningyang County in Shandong province, every year a large number of high-quality Velarifictorus micados transported to all over the country,and the local governments increase their protection to their crickets.How to identify the difference between geographical populations become the hotspot and difficulty. This study from the morphological characteristics, mitochondrial DNA COⅠgene, ribosome DNA ITS gene to identify eight geographic populations. The main conclusions are as follows:1. It can identify the geographic populations from the morphological characteristics, include antenna length, body length,body width, mandible length,mandible width, cercus length,and so on.The Taian geographic population has advantage over others in that antenna length and mandible width; the Dezhou and Jinin geographic populations has advantage over others in that body length and forewing length and cercus length.2. The research adopts the mitochondrial DNA COⅠgene as a genetic marker to analyze the eight geographic populations. Results show that the COⅠgene has no mutation among eight geographic populations, their consistency is 99.94%, no new subspecies were found.3. The study analyzes the intraspecific easy mutation of ribosomal DNA gene as the research object to analyze the eight geographic populations.The result determines the length of ITS gene is 1031 bp. There were 20 variation locuses in the ITS gene sequences,include 60% conversion mutationn and 40% transversion mutation. Based on the existed significant differences between various group, we can put rDNA ITS gene as a genetic marker.
