The Function Of Five Novel Genes During Rat Liver Regeneration Based On Transcriptome

Liver has a strong ability of regeneration. When liver is injured by operation, poisoning, infection and necrosis factors, hepatocytes can rapidly enter the G1 phase from the G0 phase which was regulated by many factors including growth factors, hormones and signaling pathways. After that, the remnant liver tissues rapidly compensated the lost weight by mitosis and cell proliferation, and recovers the function of liver, which is named as liver regeneration(LR). Liver regeneration is a very complex physiological processe, it is necessary to reavel the function of LR-related genes/proteins for the molecular mechanisms of liver regeneration. In order to explore the function of five novel genes C5ORF45, C11ORF96, C7ORF42, CXORF23 and C3ORF43 during rat liver regeneration, Rat Genome 230 2.0 array was used to detect the expression of genes at 0, 2, 6, 12, 24, 30, 36, 72, 120 and 168 h after PH. We found that the aboundance ratio of 3,886 genes were three times higher or 0.33 times lower than that of the control, which were called as meaningful expression genes, of them, 254, 9, 85, 10 and 18 genes had the same expression trend with C5ORF45, C11ORF96, C7ORF42, CXORF23 and C3ORF43, which were called as co-expression genes. Further we analyzed the signaling pathways and physiological activities that co-expression genes involved in, the results showed that 254 co-expression genes of C5ORF45 were involved in 93 kinds of signaling pathways and 37 kind of physiological activities, 9 co-expression genes of C11ORF96 were involved in 65 kinds of signaling pathways and 21 kind of physiological activities, 85 co-expression genes of C7ORF42 were involved in 67 kinds of signaling pathways and 35 kind of physiological activities, 10 co-expression genes of CXORF23 were involved in 7 kinds of signaling pathways and 13 kind of physiological activities, 18 co-expression genes of C3ORF43 were involved in 9 kinds of signaling pathways and 20 kind of physiological activities. This list of signaling pathways and physiological activities were arranged in the order of the number and proportion of their co-expression genes, which appeared in the top four signaling pathways and physiological activities of both the two methods will be considered as the function of novel genes during rat liver regeneration. The results showed that C5ORF45 played a role by participating in Tec Kinase signaling and eNOS signaling during rat liver regeneration, C11ORF96 played a role by participating in PPAR signaling and cell differentiation, cell survival, amino acid metabolism, angiogenesis during rat liver regeneration, C7ORF42 played a role by participating in Tec Kinase signaling, p38 MAPK signaling, STAT3 pathway and cellular homeostasis, coagulation during rat liver regeneration, CXORF23 played a role by participating in Paxillin signaling and cell development, cell growth, cell differentiation, locomotion during rat liver regeneration, C3ORF43 played a role by participating in Calcium signaling, p38 MAPK signaling and lipid metabolic process, DNA replication and repair during rat liver regeneration.In summary, functions of novel genes C5ORF45, C11ORF96, C7ORF42, CXORF23 and C3ORF43 during rat liver regeneration were extrapolated based on the transcriptome, which not only pointed out the direction for further experimental study on the function of novel genes but also provided data for the research of molecular mechanism during rat liver regeneration.
