Leaf color is an ideal marker to protect the purity of rice hybrid seeds. It is valuable to the research on the synthesis, structural function, genetic differentiation and development of chloroplast, as well as the synthesis of photosynthesis pigment. The thichomes that related to leaf morphology, are always independent with leaf color, previous researches mostly focus on the two traits respectively, no report about mutants with these two traits simultaneously has been seen. This essay investigates two mutants(FK3and DT48) derived from9311, which possess both white midrib and glabrous leaves, and lays the foundation for cloning genes that related to these phenotypes and further research on development of chloroplast and trichomes.Followings are the main results:1) The two mutants both experience abnormal development of chloroplasts in midribs and have much less trichomes on adaxial surface of leaves compared to the wild type9311. The decrease of chlorophyll lowered the photosynthesis rate of blade, and further influenced the agronomic traits such as seed production.2) The phenotype of glabrous leaves and white midrib were co-segregation and controlled by a pair of recessive gene; Mutants of FK3and DT48were not allelic.3) With map based cloning, the locus wmgl (white midrib and glabrous1) which contribute to the phenotype of FK3has been mapped between133550and133720(InDel maker) on chromosome4. The locus wmg2(white midrib and glabrous2) that responsible for the phenotype of DT48has been mapped between RM6301and RM5628(SSR maker) on chromosome3.
