Mango (Mangifera indica L.) belongs.to Anacardiaceae genus Mangifera L. Is one of the most famous fruit in tropical and subtropical which has economic, nutritional and social value. MADS-box transcription factor plays an important role in flowering time, floral organ development and fruit development, hs an important foundation for fruit yield and quality, so, it is important to research mango MADS-box transcription factor. In order to clone a new MADS-box gene from Mango, research its role in the development of mango flower. This study Design conserved sequences degenerate primerswas based on a variety of plant MADS-box genes, as a material with Carabao and isolated SOC1,SEP1gene by using homologous Cloning and RACE.1. The first MADS Family gene, Named MSOC1, The Accession No. is KP404094. The Coding region of MSOC1gene is733bp, and encoding223amino acids, the protein relative molecular mass25.6ku, theoretical isoelectric point is8.96. Sequence alignment and phylogenetic analysis showed that:MSOC1conserved MADS-box and semi-conservative K area, belongs to SOC1/TM3subfamily of MADS-box family. Tissue-specific expression analysis showed that:SOC1gene was expressed in various tissue sites in mango, but the expression in the stems, leaves and flower buds is high, the expression in roots and flowers is low, so presumably this gene may be play an important role in the mango flower morphogenesis and reproductive growth.2. The second MADS Family gene, Named MSEP1, The Accession No. is KP702299. The Coding region of MSOC1gene is726bp, and encoding241amino acids, the protein relative molecular mass27.7ku, theoretical isoelectric point is5.79. Sequence alignment and phylogenetic analysis showed that:MSEP1conserved MADS-box and semi-conservative K area, belongs to SEPALLATA1-like subfamily of MADS-box family. Tissue-specific expression analysis showed that:In roots, stems SEP1gene almost no expression, but the expression level increased in the leaves, flower buds, and the highest expression in the footage, so presumably this gene may be play an important role in the mango flower morphogenesis and reproductive growth.
