The capacity for nutrient transposition and absorption depend on the two Glucide transporters GLUT5 and GLUT2 in the small intestine. Caudal type hox genes Cdxa is specifically expressed in the digestive system of the duodenum to the rectum, as the transcription factor potentially regulate expression of other genes. For investigating the regulatory mechanism of Cdxa, we analyzed the hydrophobicity, PI and the structure of the protein by bioinformatics software, additionally predicted the binding sites in the 1500bp upstream of two sugar transporter genes by Modulemaster and TFbind.Through reconstructed the pcDNA3.1-Cdxa expression vector and transfected it into the IEC cell. Later, the mRNA abundance of three genes of each group was assayed using real-time PCR and the absolute quantification method. The results indicated that positive group compared with the negative group and the blank group, the expression of Cdxa is significantly increased (P<0.01), it mean that the pcDNA3.1-Cdxa is available. The expression of GLUT2 in positive group is lower than negative group and blank group (0.01<P<0.05),the result show that Cdxa played a negative transcription regulative role in GLUT2 expression. The expression levels of GLUT5was not statistically significant in different group (P>0.05).The result shows that over expression of the Cdxa play a negative transcription regulative role in GLUT2 expression.
