Stellera chamaejasme L.(Stellera, Thymelaeaceae), a perennial plant, is widely distributed in most grasslands in China. Stellera chamaejasme L. is poisonous. People may have allergic skin inflammation if touching it. It can also cause a series of disorders of degestive and nervous systems and even cause death in poisoned animals. It’s also on kind of traditional herbal medicine and has anti-baceria, pesticide and expectorant effect. Studies have showed that Stellera chamaejasme L. can produce many secondary metobilites, among which flavonoids are most. Flavonoids are the major resources for the plant’s toxicity and activity. They can poison animals, inhibit the growth of plants around, but also have anti-cancer, insecticide and anti-bacteria activity. Fungi endophytes are those who live inside the plants in its whole or part of life and don’t cause symptomous diseases. They can produce secondary metoblites similar to that produced by the plants themselves.This research isolated endophytic fungi in Stellera chamaejasme L. from 5 districts in 4 provinces. By morphological and molecular identification, the dietribution differences of endophytic fungi are described. Through fermentation, endophytic fungi producing flavonoids are selected. Research results are as follows:1. Isolation and identification of endophytic fungi in Stellera chamaejasme L.22 endophytic fungi isolated from Stellera chamaejasme L. in districts of 4 provincces belonging to 10 genus, 7 families, 7 orders and 6 classes, including Coniochaetaceae sp.、Podospora vesticola 、 Preussia intermedia 、 Preussia funiculata 、 Taifanglania sp. 、Cladosporium cladosporioides 、 Cladosporium sp. 、 Phoma moricola 、 Plenodomus henersoniae、Leratiomyces laetissimus、Alternaria sp. and Sordariomycetes sp. The number of Alternaria sp. is 9, which is the most and Alternaria sp. is the dominant gunus. Except for 1 Sordariomycetes sp. and 9 Alternaria sp., all the others are first isolated from Stellera chamaejasme L. 2. Testing for the flavonoids produced by endophytic fungi in Stellera chamaejasme L.10 fungi SC1L6-1、SC3S6-1、SC4L3-1、SC5S1-1、SC6L3-1、SC8L3-1、SC13F1-1、SC16L4-1 、 SC17L6-1 、 SY1L2-1are used. Thorough fermentation and extract of total flavonoid and tested by TLC, SC1L6-1 hyphae extract, SC1L6-1fluid extract, SC13F1-1 fluid extract and SY1L2-1 fluid extract have flavonoids. Testing for the flavonoid types and contents, SC1L6-1 hyphae extract has 2 types of flavonoids, quercetin and dihydro kaempferol, total flavonoids 0.06%. Fungi fluid extract have 3 flavonoids, quercetin, 3’,14-dimethyl-4’,11-dimethoxy-5,7-dihydroxybenzoflavanone and(+)-epiafzelechin, total flavonoids 4.60mg/L. SC13F1-1 has 2 types of flavonoids, apiginin and(+)-epiafzelechin and its content is 2.75mg/L in fluid extract. SY1L2-1 has 1 types of flavonoids, chamaejasmin A(or its isomer) and its content is 4.45mg/L in fluid extract.
