The GATA family of transcription factors is a kind of widely exists in eukaryotic transcription factors, play an important role in biological processes such as plant light reaction control, cytokines response, carbon and nitrogen metabolism. But whether it has a certain function in secondary growth is unknown now. In this paper, we cloned the GATA 12 gene which is a member of the GATA family, and research the biological function of GATA12 gene in regulation of secondary growth. The results showed that:1. Cloned Populus (Populus simonii×P. nigra) GATA 12 gene CDS fragment of 1122bp, named as PsnGATA12. Evolutionary analysis showed that the subfamily has 8 genes.2. Organization black poplar GATA12 gene expression and subcellular localization of specific analysis Tissue specific expression analysis showed that, PsnGATA12 highest gene expression in xylem, transition zone xylem and secondary xylem also had high gene expression, the roots had a low gene expression. Subcellular localization analysis showed that the protein translated by GATA 12 gene were in the nucleus localization, preliminary showed that the GATA 12 gene participate in controlling the formation of secondary growth.3. PsnGATA12 combined gene in a yeast one-hybrid experiments with cis-acting elements yeast one-hybrid technology, PsnGATA12 gene can activate multiple components of functional genes downstream of the promoter region, Show the PsnGATA12 has activation domain.4. Make the PsnGATA12 gene over expression in tobacco, transgenic tobacco plant height, leaf aspect ratio, fresh weight, dry weight, stem dry weight were significant differences between the wild-type tobacco in these biological traits. At the same time environmental scanning electron microscopy observations show that over expression of plants, transgenic tobacco secondary thickening organization. Cellulose has a length and width of the increase. The percentage content of hemicellulose compared with wild type have significant rise, The secondary cell wall synthesis and expression of genes related to the function of different degrees of change were also taken place.5. In Arabidopsis thaliana gata12mutants, the emergence of Arabidopsis thaliana in advance the reproductive growth and the phenomenon of the inflorescence stem lodging.
