High performance liquid chromatography (HPLC)is one of the main methods used inseparation science, and it plays an important role in medicine, chemistry, life science andenvironmental science. Chromatographic column is the heart of chromatographic system andthe key to develop novel chromatographic media has significant theory and practicalimportance. Because of the advantages of simple preparation and high permeability, manyresearchers have focused on the monolithic columns and the monolithic columns have beensuccessfully used in analytical chemistry.In this paper, a summary concerned with the classification and application of monolithiccolumn was introduced firstly, and the characters and disadvantages of monolith weresummarized. Then we used vinyltrimethoxysilane (VTMS) and vinyl ester resin as monomersto fabricate the hydrophilic organic-inorganic monolithic column with sonication-assist. Thestructure and performance of the prepared column were also studied. The lysozyme (Lys) wasseparated from egg white with the prepared column, and the factors affecting the retentionbehavior were also investigated.Glycidyl methacrylate (GMA) and vinyltrimethoxysilane (VTMS) were used asmonomers, organic-organic hybrid column with one pot process was fabricated, and then themonolith was modified into weak anion exchange column. The results suggested that it couldbe used to separate composite proteins and human serum albumin from human plasma, thecharacterization of the column and the factors affecting the elution of HSA were alsoinvestigated. |
