The main component of rosin is resin acids, which is the isomers of C19H29COOH. Resin acids and their derivatives have anti-tumor, anti-virus, anti-bacterial, anti-inflammatory and other biological activities, which have extensive application prospect. With the rapid development of biotechnology, biological research into a variety of drug preparation obtained greater success. Rosin resin acid and its derivatives on the biological activity of a high value of rosin resources is an important branch of research, is rapidly rising. Use of enzymes in microbial cells efficiently and selectively catalyzed rosin to obtain a highly active process of chemical composition, has broad application prospects.In this study,107 single colony fungi were isolated from pine forest humus by plate. 70 strains were selected for primary sereening, rosin for substrate, using solid state fermentation culture into them and antibacterial activity tracking. The results showed that the conversion of 16 strains have different degrees of inhibition against Gram-negative bacteria the product of bold things (E. coli) and Gram-positive bacteria (Staphylococcus aureus, Bacillus megaterium, Bacillus cereus and Bacillus subtilis).16 strains of fungi for secondary sereening, antibacterial activity form the fermentation products, and the results displayed D8 of the best strain for the transformation. Based on their morphological and molecular, D8 strain finally was identified to be Mucoromycotina, Mucorales, Mucorales incertae sedis, Umbelopsis. Then incubation conditions of D8 strain were optimized. The optimal conditions were:The initial medium pH 7.0, the culture temperature was 28.5℃, the culture time was 12d, rosin substrate addition was 2mg/mL, the inoculation amount was 10%, and the carrier diameter of cotton ball was 10mm.Under the best transformation conditions, strain D8 through the extensive transformation of solid culture rosin, the accumulation of transformation products. After alcohol precipitation method and multi-stage extraction, while the active tracking, the last of the strong antibacterial activity concentrates the ethyl acetate extract phase separation and purification of compounds.By thin layer chromatography, silica gel column chromatography, gel filtration, reverse phase silica gel column chromatography and semi-preparative high performance liquid chromatography and other chromato graphic methods, one compound was isolated from the activity of strain D8 solid-state fermentation culture of the ethyl acetate phase transformation products concentrate. This compound demonstrated remarkable antimicrobial activity in Staphylococcus aureus, Bacillus megaterium, Bacillus subtilis and Bacillus cereus.Tracking the production process of the compound by HPLC detection, we found that the strain can produce the compound itself. The addition of the rosin substrate accelerated the production of the compound, but also greatly increased the formation of the compound, that shows the rosin has induced the production of the compound. Finally, its structure was elucidated as 3,4,5-Trimethyl-1,2-benzenediol by NMR.
