Zanthoxylum schinifolium Sieb. et Zucc. is the dried ripe fruit peel Rutaceae peppers,and China is one of the largest acreage pepper in the world.Prickly ash peel can be used as acondiment, is our Chinese people very nice eating spices. In addition to peel pepper can beused as edible and medicinal, the other parts,for example, the stems, seeds, and roots, stemsand leaves can be used as medicine treatment.In recent years, Zanthoxylum schinifolium Sieb.et Zucc. has a high economic value and ecological value, growing at a rate of green pepper,more and more resports about Zanthoxylum schinifolium Sieb. et Zucc. were published, but asa kind of traditional spices, the majority research was paid close attention to its nutrition, butbut did not identify its species. RAPD (Random Amplified Polymorphic DNA) technique is atechnique of molecular methods based on the PCR technique mark, in addition to inheritingthe advantages of the outer PCR technology, RAPD technique can be studied in the absenceof the species on the basis of any molecular biology DAN polymorphisms were analyzed.Because of its high detection efficiency, small amount of sample, high sensitivity, low cost,less time-consuming series of advantages, has been widely used. In this paper, green peppervolatile oil content of the origin of the five comparative analysis and technical analysis usingRAPD genetic relationship pepper, and its results provide experimental evidence for theidentification and quality standards Zanthoxylum schinifolium Sieb. et Zucc. varieties. Themain contents and conclusions of this study are as follows:1、Pericarpium zanthoxyli schinifolii of five origin essential oil content and compositionanalysisIn this study, five steam distillation of the origin of percarpium zanthoxyli schinifoliivolatile oil and its composition by GC-MS analysis of volatile oil.The results show that theproducing area is different, the volatile oil of Zanthoxylum schinifolium Zucc prickly ashcontent is also different, there was some difference of the differences in content of origin. Inthe five regions, percarpium zanthoxyli schinifolii of Jinyang origin prickly ash peel volatileoil content in the highest, next is percarpium zanthoxyli schinifolii of Jiangjin origin. In fiveof the origin of percarpium zanthoxyli schinifolii volatile oil of main component analysis, thefive main producing area of percarpium zanthoxyli schinifolii peak group of the whole figurelooks almost the same, similar degree is higher, there are common ingredients are juniper ene,beta pinene, myrcene, limonene, linalool.2、select the optimal conditions for RAPDIn this article, Test selection of30random primers were the origin of percarpium zanthoxyli schinifolii for five performed PCR-RAPD test, selected12suitable for both fiveOrigin percarpium zanthoxyli schinifolii can be polymorphism analysis primer. And RAPDexperimental conditions were optimized. Experimental results show that the optimalparameters RAPD reaction system is:10×Taq Buffer (Mg2+Free)2.5uL, MgCl2(25mM)2uL, dNTP (each2.5mM)2uL, Taq enzyme (5u/μL)0.25uL, Primer (20pmol/μL)1uL,Genomic DNA (100ng/μL)0.5uL, ddH2O15.75uL. Cycle parameters:95℃denaturation for5min,94℃denaturation1min,38℃refolding2min,72℃extension3min,35cycles,72℃insulation10min.3、cluster analysis of RAPDA According to the obtained DNA fragments after electrophoresis map by NTSYSsoftware cluster analysis, the results show that the percarpium zanthoxyli schinifolii ofJinyang and percarpium zanthoxyli schinifolii of zhaotong belongs to the one category, thegarden empress percarpium zanthoxyli schinifolii of Jiangjin they belong to the same category,and singular percarpium zanthoxyli schinifolii of Hongya; percarpium zanthoxyli schinifoliiof Hongya empress percarpium zanthoxyli schinifolii to close some relatives, shows that thegrowth of percarpium zanthoxyli schinifolii environment climate, temperature has a certainimpact on it. The method can use percarpium zanthoxyli schinifolii can product sourceidentification.
