Optical Surface Plasmon Resonance (SPR) is a new biophysical analytical technologywhich has been used in a wide range of fields including medicine, clinic diagnosis, environmentalmonitor and food detection etc. Optical SPR biosensor has the advantage of label-free detection,real-time monitoring, and non-invasive analysis. In this thesis, coli bacillus and abscisic acid(ABA) were detected by using this optical SPR biosensor.The latest progress and research status of optical SPR biosensor technology and importanceof studying new approaches to detecting coli bacillus and abscisic acid(ABA) were firstintroduced in this paper. Then the SPR monitoring principle and method in biological detectiondomain were also recommended. Secondly, based on integrated optical SPR biosensor chipTSPRK123, biological detection instrument was constructed and different concentrations ofalcohol were monitored to test and verify its performance, then obtained the relative standarddeviation (RSD) was0.2%and the correlation coefficient of the standard curve was0.9979, whichproved that this instrument had higher performance and linearity. Finally, according tocharacteristics of coli bacillus and abscisic acid (ABA), corresponding detecting measures andexperiments were simultaneously designed and implemented to complete the whole test progress.The results demonstrate that correlation coefficient of using directing detection method tomonitor coli bacillus was-0.9851, and relative standard deviation (RSD) was6.90%. Besides,correlation coefficient of using competitive inhibition method to monitor abscisic acid(ABA) was-0.998, and relative standard deviation (RSD) was4.13%.
