Panax Notoginseng was known as an important crude drug in traditional Chinese medicines. It was widely used to cardio-cerebrovascular diseases, liver disease and cancer treatment. A great deal of pharmacology researches discover that the saponins of panax notoginseng was the effective components. In order to clarify the chemical foundation of saponins composition, In this paper, the preparation, isolation and identification of the saponins of panax notoginseng was researched.In the article, the TLC and HPLC was used as the quantitative and qualitative analysis. The normal phase eluant was optimized by using TLC, and the preparative C18eluant was optimized by using of C18chromatography technology. The saponins of panax notoginseng was isolated and purified by using of macroporous resin HPD100, silica gel and reversed-phase C18; The compound was characterized by LC-UV and LC-MS.The results indicated that the analysis conditions of TLC was n-butanol:ethyl acetate:water=4:1:5, and the analysis conditions of HPLC was gradient elution of acetonitrile-water. The macroporous resin HPD100was used as the first column chromatography to get6distillates L2~L7.The normal phase eluant was dichloromethane:methanol=100:8~100:50, then silica gel was used as second column chromatography to get14more pure distillates Y1~Y14.The elution condition of preparative C18was methanol:water=65:35, and the14saponin monomers were purified by C18, then9kinds of saponin LY1~LY9were obtained. They were identified as gensenosides Rg1,Re,Rb1,Rc,Rb2,Rb3,and notogensenosides Fa,Fc,,Rd. The paper summed up the chromatographic behavior of four saponins LY1、LY2、LY3、 LY9on HPD100,silica gel and C18. A further research of the flow discipline was needed.
