Corn is an important economic crops due to widely grown in our country,especially in northern China is gradually expanding its acreage. Our country is the second largest producer of corn in the world. Natural pigment rich in corn renewal as an important source of natural antioxidants can protect eyesight and prevent senile macular degeneration and cataracts, and other important health functions. As widely used in health food processing, therefore it has been a high application value and development prospects.This test was to research the extraction and purification technology of yellow pigment from corn protein powder of local feed mills as raw material, it was identified that the microcapsule technology can protect the stability of maize yellow pigment.This provides a basis for the development and application of maize yellow pigment.The main findings are as follows:1.Optimization of extraction processes of maize yellow pigmentAccording to single factor experimental results, The optimum parameters of the ultrasonic microwave assisted extraction process of maize yellow pigment were determined by rotatable central composite experimental design and response surface analysis are:ultrasonic time was70min, ultrasonic power was70w, microwave time was80s, microwave power was450w. Under this conditions,maize yellow pigment extraction yield reached97.022%which was close to the predictive value of97.022%.2. Purification and Preliminary identification composition of maize yellow pigmentBy comparing the adsorption rate and the desorption rate of10kind of macroporous resins on purifying anthocyanin from black rice, the macroporous resins with best purification effect was HZ816. The optimum purification conditions were determined by adsorption and desorption test of static and dynamic:adsorption time:2h. adsorption pH:3.0, desorption time:2h, desorption solvent:90%ethanol, the adsorption flow rate was:1.6ml/mL, desorption flow rate was1.5mL/min and the sample concentration:0.04g/mL.The pigments were judged for Carotenoids by UV and IR.3.Optimization of microcapsules processes of maize yellow pigmentAccording to single factor experimental results. The optimum parameters of process for the microcapsules were determined by the orthogonal method are:pigment and beta-cyclodextrin ratio1:6, beta cyclodextrin concentration of10%. the grinding of50min, under this condition, the embedding rate of maize yellow pigment microcapsule can reach62.1%.4.The stability of maize yellow pigment The stability of maize yellow pigment were studied under the common condition.we got the following conclusions:(1)The stability of maize yellow pigment is very good in acid to weak alkali conditions, but the stability was destroyed under alkali condition.(2) The heat resistance and the light fastness of maize yellow pigment were poor, so it should be in low temperature and avoid light.(3) The oxidant has big influence on maize yellow pigment,but the influence of reductant is small.(4) Fe3+, Ca2+, Cu2+, Al3+has big influence on maize yellow pigment, the influence of Fe2+Zn2+can ignore, and Na+, K+, almost has no influence.(5)Citric acid, starch, sodium bicarbonate and other commonly used food additives will have little impact on maize yellow pigment, and vitamin C can protect maize yellow pigment. |
