Glycosaminoglycans from Urechis unicinctus was extracted and separated, and chemical structure and composition of purified glycosaminoglycans was analyzed. On this basis, the biological functions of glycosaminoglycan were studied.Optimization of extraction conditions for glycosaminoglycan from U. unicinctus by response surface methodology:The enzyme dosage, enzymolysis time, and alcohol precipitation concentration as independent variables, the extraction rate as the response value, and the extraction technology of glycosaminoglycan from U.unicinctus was optimized via response surface methodology on the basis of single-factor experiments. The extraction rate of glycosaminoglycan was affected in the following order:concentration of alcohol precipitation>enzymolysis time>enzyme dosage. The optimal technological conditions for the glycosaminoglycan extraction were as follows:alcohol precipitation concentration,75%, enzymolysis time4h, and enzyme dosage,0.4%.The average content of sulfate, uronic acid and amino sugar in the glycosaminoglycan were30.26%,25.25%and7.58%respectively using chemical method; Infrared spectrum of the glycosaminoglycan was analyzed by KBr compression method, showed that glycosaminoglycan contained characteristic groups such as-OH (3418cm-1),-COO-(1642cm-1),-SO2(1415cm-1) and C-O-C (saccharide ring,1034cm-1) etc; within the range of0.1-0.5mg/mL, glycosaminoglycan had a strong scavenging effect on hydroxyl radicals, and scavenging rate was above80%, but had a weak scavenging effect on superoxide anion radical; In vitro experiments, the glycosaminoglycan could obviously extend the PT and APTT of big ears rabbits, slightly prolonged TT, there may be a anticoagulant way different from the heparin; Diabetic mice were induced, the hypoglycemic effect of glycosaminoglycan from Urechis unicinctus on diabetic mice was investigated. Results demonstrated that glycosaminoglycan could significantly reduce blood glucose concentration, HOMA-IR, AUG and liver MDA content in diabetic mice (P<0.05, P<0.01). In addition, it significantly enhanced liver SOD and GSH-Px activity (P<0.05, P<0.01), as well as liver GCK activity and hepatic glycogen levels. It also had good curative effect on diabetic mice, showing promising prospects for market development.
