In recent years,the plant polyphenols was researched and developmented more and more bydomestic and foreign experts.Plant polyphenols were plant complex phenolic secondarymetabolites,it had a variety of physiological activity, such as inhibition of inflammation, tumor,virus and antioxidant.Inonotus obliquus was a kind of wild medicinal and edible fungi,the fruitbodies contains a variety of bioactiv substances,the main active substance for the antioxidant waspolyphenols.In order to make full use and improve the comprehensive utilization of Inonotusobliquus,this topic taked Inonotus obliquus as raw materials,based on single factorexperimeng,optimized the process conditions of extraction for polyphenols from Inonotusobliquus.From5kinds of macrporous resins were screened in a suitable for purification ofpolyphenols from Inonotus obliquus,and determined the optimum process conditions.Thecomponents of polyphenol from Inonotus obliquus were inferred.Finally, the antioxidant activityof the purified and crude extract of polyphenol from Inonotus obliquus were compared.Thistopic made a systematic research on the polyphenols of Inonotus obliquus, in order to providesome support for the development of Inonotus obliquus.Below were the main research contentsand conclusions.(1)Studied the process conditions of microwave extraction and solvent extraction ofpolyphenol from Inonotus obliquus.With the results of single factor test as the reference, usingthe orthogonal experiment optimization process conditions of two kinds of method for extractingpolyphenol from Inonotus obliquus.The results of the study showed that, the ratio of material toliquid was1:20, ethanol volume fraction at50%, extraction temperature at60C, time at80min,under the conditions of extraction, the extraction rate of polyphenols was2.633%in theexperiment of solvent extraction for polyphenol from Inonotus obliquus;The ratio of material toliquid was1:50, ethanol concentration at50%, microwave power at560W, time was180s, underthe conditions of extraction, the extraction rate of polyphenols was3.186%in the experiment ofmicrowave assisted extraction of polyphenol from Inonotus obliquus.The microwave assistedextraction is an effective and rapid method for extracting polyphenols from Inonotus obliquus.(2)Studied the process conditions of the macroporous adsorption resin separation prificationfor polyphenols from Inonotus obliquus.From5kinds of macrporous resins were screened in a suitable for purification of polyphenols from Inonotus obliquus was D101by staticadsorption,desorption experiment and the dynamic and static adsorption experiment,and theprocess conditions for purification of polyphenols from Inonotus obliquus was determined.Theresearch results showed that,the optimum purification process of polyphenols from Inonotusobliquus was the glass column at40*1.6cm,the concentration of sample was1.0mg/mL,thesample speed was1.0mL/min,the eluent was70%ethanol solution,the rate of elution flow was1.0mL/min.Under the optimum conditions of purification was the purified of polyphenols fromInonotus obliquus that purity was improved from16.52%to49.77%,increased3times.(3)Components of polyphenols from Inonotus obliquus substance were identified preliminarilyby using the technology of HPLC-ESI-MS.The research results showed that,the mechanism ofpolyphenols from Inonotus obliquus contains9substances, their names wereLeucoanthocyanidins,Astilbin,Curculigoside,Quercetin-3-O-glycoside,Catechin7-xyloside,CucurbitacinⅡb,Tangeretin,Catechin,Rutin.(4)Studied the antioxidant activity of polyphenols from Inonotus obliquus extracts andpurified.Using DPPH free radical scavenging activity and hydroxyl radical scavenging ability toevaluate the antioxidant activity of polyphenols from Inonotus obliquus.The research resultsshowed that,the IC50value of Vc was6.29μg/mL,the IC50value of polyphenols from Inonotusobliquus purification was6.82μg/mL,the IC50value of polyphenols from Inonotus obliquusextracts was10.73μg/mL in the DPPH free radical scavenging assay.The IC50value of Vc was1.10mg/mL,the IC50value of polyphenols from Inonotus obliquus purification was1.54mg/mL,the IC50value of polyphenols from Inonotus obliquus extracts was2.49mg/mL in thehydroxyl radical scavenging assay.According to the experimental results,the polyphenols fromInonotus obliquus purified compounds had antioxidant activity,second only to the antioxidantcapacity of Vc,but was much stronger than the crude extract.
