In the past decades, nanomaterials have become an import research focus withthe increasing attention and interests from both scientific research and industrialapplications. With the rapid academic progress and technical developments,nanofibers have successfully assessed into many aspects in our daily life. Electrospunfibers have been promisingly applied in such many medical applications as carrier forcontrolled release of drugs, tissue engineering and wound excipients. As the novelcandidate for controlled release of drugs, the electrospun fabric membrane, formed bynanofibers spinning under the electrical fields and the fast evaporation of organicsolvents, resulting homogenous loading of drugs in the fibers, would release the drugsloaded in a controlled manner based on the biodegradation of polylactic acid matrix.Oral cancer has become a frequently pathogenic cancer in the recent years, andtraditional therapy is still mainly based on surgery aided by chemotherapies. Whilethe strong chemical side effects and hardly balanced drug concentration in vivo hasbeen puzzling problems. This thesis work will focus on a controlled release systemfrom electrospun nanofibers as drug-carriers based on the membrane fabrication andtheir interaction with CAL-27oral cancer cells. The following results have beenobtained:1.Effects of various concentration of spinning solutions on meso-scaledmorphology of nanofibers have been investigated. Cisplatin has been successfullyloaded into nanofibers with PLA matrix as the matrix by means of electrospinning. Ithas been proved that the drug release has been performed in an effectively controlledmanner with strong and sustainable effects when comparing the conventional drugsinteracting with CAL-27oral cancer cells.2.Through the introduction of chloroquine as autophagic inhabitors, the drugsensitivity of oral cancer cells on cisplatin drugs has been dramatically imporved.With fractionated dosing in the nanofibers loaded with chloroquine and cisplatin, thehealing effect on previously cisplatin loaded nanofiber membrane has been strengthened dramatically, better curing results, much lower side effects, whencomparing with only cisplatin laded nanofiber membrane.
