Angiogenesis is the sign of tumor growth, and anti-angiogenic drugs can reduce the blood supply of tumor and inhibit its growth. Anti-angiogenesis therapy has become a major field of study for tumor targeted therapy. It is very critical to accurately assess angiogenesis or injury situation. At present, a variety of imaging techniques used can not detect angiogenesis or blood vessel impaired from the molecular level. Therefore, looking for a molecular detecting method and appling it to the research of blood vessel generating or damaged, will be great significant to the clinical treatment of cancer.In this paper, for the above problem, Raman spectroscopy was applied to the study of anti-angiogenic drugs on the vascular injury. The non-shell chick chorioallantoic membrane (CAM) was used as the experimental model, and 785 nm semiconductor laser was used as the excitation light source. We accessed and relatively analysised the Raman spectra of the anti-angiogenic drugs before and after acting on vascular injury, and preliminary investigated the application of laser Raman spectroscopy in tumor anti-angiogenesis therapy and anti-angiogenic mechanism.First of all, the research background and the significance of the selected topic were elaborated, and the basic principle of Raman spectroscopy and its research progress in tumor anti-angiogenesis therapy and anti-angiogenic mechanism of drug action at home and abroad, was briefly introduced.Second, experimental model of the shell chick chorioallantoic membrane (CAM) was established and Raman spectroscopy was used to study the vascular growth of the chick embryo chorioallantoic membrane and anti-angiogenic drugs on chick vascular injury;Finally, the Raman spectra of the variant nasopharyngeal carcinoma organization before and after the drugs were studied.The results showed that:(1) Raman spectroscopy could be applied to the chick chorioallantoic membrane vessels reseach; (2) Raman spectroscopy could analyze the mechanism of anti-angiogenic drugs from the molecular level. This would provide a new research method for the research of anti-angiogenic therapy; (3) The non-shell embryo, in which allogeneic tumor was planted simply, was a good animal model of studying the Raman spectra of nasopharyngeal carcinoma with and without drugs acting in vivo.
