As an important part of the mitochondrial transcription, Mtfl (Mitochondrial transcription factor 1) is encoded in the nuclear, assemblied in the cytoplasm and finally transported into the mitochondria in Schizosaccharomyces pombe. Mtfl can regulate mitochondrial transcription by recognizing the complex of RNA polymerase and promoter and initiating the transcription. It's currently unknown how the Mtfl enters into the mitochondria and plays role. We found the interaction of Hsp60 and Mtfl in vivo by Tandem Affinity Purification and MALDI-TOF MS. We proved the interaction of them in vitro by GST-pull down assay with the recombinant GST-Mtfl and MBP-Hsp60. We got the association and dissociation curves and measure the affinity constant by SPR. assay. ETB is the specific inhibitor of Hsp60. As results shwoed:ETB could destroy the binding of Hsp60 and Mtfl. ETB inhibited the growth of the wild type 972 cell. ETB had no effect on mtDNA but repressed the transcription of mitochondria genes. We concluded that ETB inhibited the Hsp60 activity and disrupted the interaction between Hsp60 and Mtfl. This could affect the mitochondria gene transcription since the transport of Mtfl from nuclear to the mitochondria was blocked.
