The active targets of the aneugens in the cells were not only chromosome and DNA but also all kinds of mitosis apparatuses (such as centriole, kinetochore, microtubule of spindle etc ),centromere and cell membrane. It was much more important for us to acknowledge that all kinds of different aneugens would act on different targets and that some aneugens even acted on more than one target ,that is to say ,their targets was not sole Accordingly , in order to afford more scientific reasonable principle for protecting the health of mankind from adverse things ,we should immerse in finding out the active mechanism of all kinds of different aneugens in order that make the pathways of the specific aneugen inducing the aneuploidy clear. Based on the above laws, tubulin was separated and purified from the nascent porcine brain by two steps of polymerization and depolymerization reaction .And then we set up a set of balanced reactive system of the tubulin polymerization and depolymerization .Some approved aneugens (Trip terygiuwlIypog]aucum (]evel)Hutch, colchicine, thiabendazole, a tropine sulfate,d-ubocurarine) was added into the reactive system respectively and the changes of their absorption values were taken down during different periods under the conditions of 37 0C and 350nm .These datum were compared with that of positive control (colchicine) and solvent control (double distilled water or DMSO) at the same time point .we made use of the comparative results to assess the influence of these kinds of aneugens on the polymerization of tubulin in vitro and then draw a conclusion on the mechanisms of the aneupoidogens inducing aneuploidy on the side The result showed: (1)The three concentrations of positive control (COL) (0. 0025mM, 0. 0050mM, 0. 075mM) strongly inhibited the polymerization of tubulin in vitro and their absorption values were notably lower than that of the solvent control at the same time point . (2)The highest concentration group(l.92mg/ml)among the three concentrations of THH inhabited the polymerization of tubulin as COL did and the relationship between inhibitive response and concentrations appeared positive correlation clearly (3) The dose concentrations of atropine sulfate(ATR) and dę”¾ubucararine(dę”¾UB) appeared the same inhibitory response on the polymerization of tubulin and the relationship between inhibitive response and concentrations was similar as that of TUII. (4)Compared with its own solvent control (DMSO), Thiabendazole (TBZ) showed apparent inhibitory response on the polymerization of tubulin and the relationship between inhibitive response and concentrations also took up positive correlations. The results indicated that THH,ATR,d-TUB and TBZ showed inhibitory response on the polymerization of tubulin in vitro clearly and positive correlations between inhibitive response and concentrations. We suggested that the four approved aneugens possibly induced aneuploidy by using the pathway of inhibiting the polymerization of tubulin...
