The refractive index of biological tissues is one of the important contents in basic study of tissue optics, which is valuable to setting up a tissue optics model and describing the light transporting process in biological tissues. Directing at handling the refractive index of biological tissues, surface plasmon resonance technology (SPR) and elliptical polarization method were adopted here to research the testing problems in experiment.Surface plasmon resonance technology was introduced into the testing study of the refractive index of biological tissues, whose testing theory is discussed theoretically. Considering the environment and other influences, an Au film of 50 nm was selected as the wafer film. Relative experimental setup for SPR method were designed and established. The experiment was carried out with distilled water, grain alcohol and IntralipidTM , whose results had a good consistency with the data obtained using traditional methods. As a new testing method to refractive index of biological tissues, surface plasmon resonance technology has special merits and an important application in future. The research results have some reference values to the real-time test of refractive index of true biological tissue.Then, the complex refractive index of true biological tissues was studied by the elliptical polarization method. Some typical biological tissues, such as porcine muscle, porcine liver, porcine pancreas, were measured by such method. Experimental results agreed with other reports well, which demonstrated the elliptical polarization method was an effective way for measurement of biological tissues.Surface plasmon resonance technology and elliptical polarization method are both the groping works for the refractive index of biological tissues. Other problems such as software analysis and sample preparation are necessary to be studied in future work.
