The behavior of K~+ channels current in onion pellicle cell is received by patch clamp system. Injecting different quantity Cu to the resolution out of the microelectrode, the behavior of K~+ channel current can be used to investigate the characteristic of K~+ channel impacted by Cu in onion pellicle cell.This paper contains mainly as follows:1. The calibration the solution components in and out of the microelectrode, infiltration pressure. The components in and out of the microeletrode should be same with the solution in and out of the cell. Change the quantity of the D-mannitol to change their infiltration pressure, make them close as possible as you can.Get the connections between bioplasm quality and enzyme concentration, enzyme effect time, centrifugal velocity, centrifugal time and centrifugal times. Cell wall is an obstacle to clamp, we need to take off the cell wall with enzyme. And the bioplasm quality is one of the most important factors to get the Giga seal. The above factors decide the bioplasm quality, try to get their connections and extract the best data in order to finish the next step.2. Get the impact of current through ion channel after dealing with the copper element. Change the solution concentration of copper element in and out of the microeletrode, then get their connections. With the Origin7.0 software, the data can be analyzed to the influence of K~+ channel by Cu element.It is the K~+ channels that are one of the most widely distriuted ion channels of organisms, so it is important to investigate the impact and analyse the reasons of this impact when the environmental factors changed. People can realize the ion channel from microcosmic state.Through recording the changes of ion channel current, the state of ion channel can be judged, so the plant reactions can be analyzed.
