Objective:Wujiwan composed of three herbs such as Coptis,Evodia rutaecarpa(Juss.)Benth,Paeonia lactiflora Pall. The major functions of Wujiwan are liver-fire attacking stomach, stomach-ache, acid requrqitation, bitter taste and diarrhea. Ultra-fine power technology for compound preparation can enchance the content of the effective ingredients in pharmaceutical preparations, dissolution rate. In this paper,Ultra-fine powder techonlogy applies to preparation of Wujiwan with the view to compare the pharmaceutical technology and the content of majoy effective ingredient with general Wujiwan, apply experimental data for the manufacture.Methods:By comparing the pulverization technology of Coptics in Wujiwan,to choose the best pulverization technology.By comparing pharmaceutical technology, dissolution of effective ingredient and the content of majoy effective ingredient between the ultra-fine wujiwan prepared by ultra-fine powder technology and the general wujiwan, apply experimental data for the manufacture.1. Pulverization technology research:2010 Chinese Pharmacopoeia regulation that the cotent of Wujiwan take the berberine hydrochloride as standard.Choose the berberine hydrochloride as index, inspect the particle size, extraction, and time on the content of berberine hydrochloride effect,Choosed the best pulverization technology research by orthogonal experiment.2. pharmaceutical technology:By using Beili grinder to crush crude drug of the prescription, made pills,and determine the content of water in the pills. 3. The experiment to detemination the dissolution of berberine hydrochloride in wujiwan pills:Dissolution rate by pulp method, fitted with Weibull model and do statistical analysis with t50,tD,and m.4. The determination of content in the compound ingredient:(1) Berberine hydrochloride as a target index, determinated with HPLC and compared with general wujiwan. (2) Paeoniflorin as a target index, determinated with HPLC and compared with general wujiwan. (3) Evodianmine and Rutaecarpine as a target index, determinated with HPLC and compared with general wujiwan.Results:Through comparative study of comparative in dissolution on the general wujiwan and ultra-fine wujiwan,show that the differences of t50 and tD between the ultra-fine wujiwan and the general wujiwan have statistical significance; The t50 and tD of berberine hydrochloride in the ultra-fine wujiwan were lower than the general wujiwan, t50 of ultra-fine wujiwan was 2.90552±0.35, tDwas3.41692±0.64 and the general t50 was 20.52933±0.43, tD was 20.49374±0.73. Through compare the content of majoy effective ingredient between the ultra-fine wujiwan prepared by ultra-fine powder technology and the general wujiwan, the content of berberine hydrochloride, paeoniflorin, evodianmine and rutaecarpine were significant differences.Conclusion:By using ultra-fine powder technology in the preparation of wujiwan, it could improve the extract, the active ingredient rate of extraction and in dissolution. Through the comparative content determination of berberine hydrochloride, paeoniflorin, evodianmine and rutaecarpine in ultra-fine wujiwan were higher than general wujiwan. The study has provided experimental data for application of ultra-fine powder technologh in ordinary Chinese medicine.
