| In this paper, Lactobacillus reuteri IMAU10281, a potential probiotic lactic bacteria, was chosen as the research object, optimization of medium and culture condition were studied to provide basic data for the high cell density culture of Lactobacillus reuteri IMAU10281. The main research conclusions are as follows:In order to increase the viable bacteria counts of Lactobacillus reuteri IMAU10281, this study based on the culture medium MRS, all ingredients include the carbon sources, nitrogen sources, total carbon and nitrogen and carbon-nitrogen ratio, buffer salts, trace elements and growth factors were screened and their concentration was adjusted, then using the response surface design to optimize the composition proportion, forming the optimal medium of Lactobacillus reuteri IMAU 10281:glucose 6.92 g/L, sucrose 6.92 g/L, lactose sugar 6.92 g/L, soy peptone 25.96 g/L, casein peptone 12.98 g/L, Na2HPO4 32.46 g/L, citric acid 3.88 g/L, MgSO40.2 g/L, MnSO40.08 g/L, Tween-801.0 g/L, L-cysteine hydrochloride 0.2 g/L. When cultivated in the optimum medium at 37℃ for 12 h, the viable count of fermented broth can get to 6.30x108CFU/mL, which was 6.81 times as many as before (9.25×107CFU/mL). |
