A new extraction technology of green and efficiency——aqueous two-phase system is used to leach alkaloids from rhizoma coptidis crude extract. (1) Prepared crude extracts from rhizoma coptidis alkaloids by acid-water immersion method, and set aside. (2) Using polyethylene glycol (PEG) and ammonium sulfate aqueous two-phase system to extract rhizoma coptidis alkaloids. Under the condition of pH=6.0 and temperature 20℃, through the study on the composition of aqueous two-phase systems:the molecular weight and concentration of PEG, the kinds and concentration of salt, which are influenced the extraction of berberine, coptisine, palmatine and jatrorrhizin in aqueous two-phase system. Then combined with ultraviolet spectrophotometry to detect out the extraction craft of four kinds rhizoma coptidis alkaloids when the extraction efficiency is the highest. When PEG1000 mass fraction 20%, ammonium sulfate mass fraction 16%,22%,16%,20%, the highest extraction efficiency of berberine, coptisine, palmatine and jatrorrhizin are 99.79%, 98.04%,99.96%,99.39%, respectively. (3) Using aqueous two-phase system to extract the alkaloid of rhizoma coptidis crude. By the orthogonal experiment to determine the optimum extraction conditions:pH=6.0, temperature 20℃, PEG1000 mass fraction 22%, ammonium sulfate mass fraction 15%, the highest extraction efficiency is 99.67%.
