Collecting test samples koumiss from Mongolia and isolated and identified morphologically, screening the best quality strains from the koumiss, the fresh mare’s as culture medium,Optimization of fermentation conditions that producing antimicrobial substances, while antibacterial substances were extracted, its antibiotic activity was detected by cylinder-plate method.Density of the target strains, concentration of agar, culture time have effect to computation. Select the best experimental conditions can improve the accuracy of antibacterial experiments.11 lactic and bacteria and 14 yeasts strains, isolated from 15 koumiss samples of Mongolia region, The morphologically and physiologically identified of 11 strains lactic and bacteria results was 3 genus,2 of 11 strains was Enterococcus,3 identified as Lactococcus,6 identified as Lactobacillus.14 yeasts identified as Pichia、Brettanomyces、Endomyces、Saccharomycodes。Screened two strains of lactic acid bacteria inhibitory ability Y1 and 10A1, And two strains of lactic acid bacteria and 14 yeasts mixed culture, compared to the single lactic acid bacteria fermentation antibacterial activity. Screening lactic acid bacteria and yeast combination 10A1-J10A1 and Y1-JX2 which can significantly promoted antibacterial substance produced. Combination metabolites Y1-JX2 were more sensitive to protease, Protease can significantly reduce their inhibitory activity. Screening kumiss fermentation conditions. Single factor experiment and response surface methodology to optimized the fermentation conditions. Get the fermentation temperature 33℃, inoculation was 8%, the proportion of vaccinated was 5.6:2.4, the fermentation time was 84h and initial pH=7 were the best fermentation conditions. Inhibition zone diameter was 17.70mm. The actual value consistent with the predicted value after response surface optimization experiments, Alcohol within the range of 1%-2%vol, total acidity was 80-120° T. Total sugar 30.2-56.8g/L. Experimental results show that,15%to 20%(NH3)2SO4 can isolated some non-protein inhibitory substances, 60% saturation can be isolated the most of the inhibitory substances. Antibacterial substances mainly in the dialysis bag. By electrophoresis, which molecular weight was about 70kDa. Separation methods still need to be studied.
