Betulin used in this paper is secondary metabolites from natural plant extracts in the bark, and betulin is a pentacyclic triterpenoids with anti-tumor, anti-bacterial, anti-HIV, anti-virus and other biological activities. And according to recent studies show that thiosemicarbazone metal complexes also have a lot of good biological activities on antitumor, antibacterial and other biological activities. So this paper, the basic skeleton of betulin, in its C-28 introduced thiosemicarbazone metal complexes, both the joint of them in order to improve the purpose of anti-tumor and anti-bacterial of biological activities.In this paper,the betulin raw extract from the birch powder. First, we protect 28-OH with triphenylchloromethane,and re-introduced at 3-OH with ester groups,then on the 28th position after deprotection, oxidation, introducing thiosemicarbazone, and finally to the compound (6) synthesized 3-O-acetyl-betulaldehyde thiosemicarbazone ligands with various transition metalion complex and get the final products: 3-O-acetyl-betulaldehyde thiosemicarbazone metal complexes. The metalions of the final product synthesized in the paper are copper, silver, cadmium three metalions, and the synthesis of metal complexes were through IR,1H NMR, TGA, MALDI-TOF mass spectra and elemental analysis. A series of tests by the above test results show that these ligands structure:copper and cadmium complexes are double ligands, the complexe of silver is double ligand with two water molecules. And the ligand and three kinds of betulaldehyde thiosemicarbazone metal complexes were determined by MTT and HIF-1 method were tested in vitro anti-tumor activity, as well as Gram-positive and Gram-negative bacteria antimicrobial activity test the bacteria activities.
