Diosgenin is the important raw material for the production of steroid hormone drugs with a variety of biological activity. Dioscores zingiberensis C.H. Wright contains the most dioscin in the world. Dioscin desugar to obtain diosgenin. The traditional production process of diosgenin use high concentrate sulfuric acid or hydrochloric acid, and lead to a large number of serious pollution to the environment. The purpose of this paper is to develop a cleaner production process of diosgenin and achieve full use of the resources at the same time. First of all, we study on the microbiological method, enzymatic method and compound method to product diosgenin, successfully obtained a Compound enzyme cleaner production process. Then this process is optimized and explored its mechanism to pave the way for realizing cleaner production process of industrialization. The main research contents and results of this paper are as follows:1.According to the rate of diosgenin,2 kinds of microorganisms (Aspergillus niger/Aspergillus oryzae); 11 kinds of combination process of 9 kinds of commodity enzyme; and compound method process were compared, selected a Compound enzyme cleaner production process, and put forward the three steps conversion theory. One is the destruction of starch shielding, exposure of dioscin; two is breaking of cellulose bonding to free dioscin; three is the dioscin change into diosgenin.2. Optimizing the process conditions, the best reaction conditions were found in the pH 5.0,50℃ under the conditions of enzymatic reaction 48h, joined the p"-glucosidase enzyme reaction of 36h, the process of diosgenin yield is 31.68mg/g, and production of glucose 230mg/g. For a greater degree of increase diosgenin yield, in the original process of composite acid solution, the new process increased the diosgenin yield to 128%, and reduced 70% acid.3. Through the study of 6 kinds of dioscin distribution, enzymatic conversion pathway and TGA, FTIR characterization of Compound enzyme production process, discussed the mechanism of Compound enzyme production process. In the process of Compound enzyme method, cellulose enzyme played a damage starch cellulose shield, cut off the winding, full free dioscin role.β-glucosidase change dioscin into diosgenin. Study on the distribution of dioscin said, there are 53% hydrophobic and 47% hydrophilic saponins in natural diosgenin. Compound enzyme action makes the shortening of sugar chain of saponins, even more hydrophilic into hydrophobic dioscin. Therefore the solid slag contains more free diosgenin, mixed acid solution will further transformation of solid residue of dioscin to diosgenin, and improve the yield of diosgenin.At present, there are many explore on bioconversion process of diosgenin and recombinant enzyme transformation experiments, but little explore on commercial diosgenin enzymatic conversion. In this paper, for the first time by using commercial enzyme completely change yellow ginger into diosgenin, achieving full utilization of plant resources. And put forward the concept one pot method, it is a simpler method and provide a theoretical support to the industrialization.
