Persistent organic pollutant perfluorooctane sulfonate(PFOS) has chemical stability, bio-inert, and hard- volatility, and it is very difficult to degrade in ecological system. Now it has become one of the most serious contaminants which threaten the environment and public health, as it is detected in environmental medium in the world like water, atmosphere and sediment as well as in liver and serum in human body and animal. By taking the scenedesmus obliquus and Danio rerio as model organism, the essay explore the best way of algae cell disruption. And the essay also shows the effects of PFOS on acute toxicity of scenedesmus obliquus and on some physiological and biochemical indexes like soluble protein, SOD, CAT, MDA of scenedesmus obliquus, ALP, LDH, Ach E, visceral mass(ALP, SOD, CAT, MDA) and musc Le(ALP, SOD, CAT, MDA) in the head of zebrafish. The results are in the following:1. After one round freeze-thaw and 25 min ultrasonication, The algae cell can be completely broken.2. In the acute toxicity test, Method of cell counting, spectrophotograph, and chlorophyll a analysis are used to reflect the change of algae density and the corresponding 96-EC50 were 75mg/L, 98mg/L and 95mg/L respectively.3.From the experiment, PFOS will restrain the protein content of scenedesmus obliquus, but the difference is not obvious(P>0.05). The activity of SOD will be restrained in the first and then abducted by high content PFOS. at 48 h, the value of SOD is striking higher than the control group(P < 0.05) by the high content PFOS(120mg/L). The PFOS has induction on the vigor of SOD. With the PFOS in high concentration(120mg/L) at 48 h, the CAT value is striking higher than the control group(P<0.05). The content of MDA is showed abducting first and then restraining with the effect of PFOS, but it has no obvious difference with the control group(P>0.05).4. The ALP in the head of zebrafish will appear stress reaction in high concentration(5mg/L, 10mg/L) PFOS at 0.5h and 1h. The vigor of ALP will be restrained and back to normal by autogenous regu Lation. The vigor of LDH does not appear stress reaction, but abduction in mid&low concentration instead. There is no obvious significance in high concentration in 0.5~1h. The vigor of Ach E begins stress reaction at 0.5h, the mid and high concentration suffers striking(P<0.05) and extremely striking restraint(P<0.01), then at 96 h, the low(1mg/L) and high(10mg/L) suffer remarkable abduction(P<0.05).5. The vigor of ALP of zebrafish visceral mass in high concentration exposed group rises entire Ly by suffering abduction. The vigor of SOD in different concentration group rises first, then goes down and tends to stable in the end. With the effects of PFOS, the SOD is abducted observably at 0.5h and 1h. Although it goes down, it does not suffer remarkable restraint. The vigor of CAT has great difference with the control group(P<0.05) at 0.5h and 1h, as the stress reaction abducted in low concentration. Then it is observably abducted(P<0.05) in the 48 h 5mg/L concentration. The rest has no significant difference(P>0.05). The MDA in the visceral mass responses the most obviously by the effect of PFOS. During the 0.5~1h in stress reaction, the content of MDA in mid&high concentration group is increased obviously, which has an extremely difference with the control group(P<0.01). Although it comes back to a certain level, the content increases during 12~24h, which has an extremely difference with the high concentration group(P<0.01).6. The vigor of ALP in zembrafish muscle responses obviously during 0.5~12h. Except the 3h, the vigor of ALP in other mid&high concentration group is restrained. However, the PFOS has little effect on the vigor of SOD. The infected group has no obvious difference with the control group(P>0.05). With the effect of PFOS, the content of MDA in stress reaction appears abduction in low concentration, and restraint in high concentration at 0.5h. it will recover to a certain level, but the restraint appears again in mid&high concentration group at 12 h, 48 h and 96 h.
