The HPLC fingerprinting of ke-chuan-shun pills was established with column Agilent SB-C18(5μm, 250×4.6mm). The mobile phase was ACN(A)- 0.1%PI/H2O(B), and the gradient elution was applied in chromatographic separation: 0min, 5%A; 30 min, 30%A; 120 min, 95%A. The flow rate was 1ml·min-1, the detection wavelength was 225 nm. The column temprature was 30℃, and the injechtion volumn was 10 ul.3. The GC-MS fingerprinting of ke-chuan-shun pills was established with quartz capillary column HP-5MS(5% Phenyl Methyl Siloxane, 30.0m×0.25mm×0.25μm). The initial column temperature was set at 70℃ and maintained for 1 min, then programmed at 20℃·min-1 to 190℃and maintained for 2 min, then 17℃·min-1to 225℃, then 5℃·min-1 to245℃ and maintained for 1 min, then 2℃·min-1 to 265℃ and maintained for2 min. Splitness injection was conducted and helium was used as carrier gas of 1ml·min-1 flow rate, The spectrometers were operated in electron-impact(EI)mode, then scan range was 50-550 amu, the ionization energy was 7 e V. The inlet and ionization source temperature were 270 and 230℃ ℃, respectively. |
