Objective:Erythritol is a new type of sugar alcohol sweeteners, in recent years, the study found that it has inhibition of bacteria. But mainly concentrated the study in the erythritol on streptococcus and porphyromonas gingivalis, the reports about the effect on the growth and acid production of lactobacillus who is one of the main cariogenic bacteria in the mouth were rare.Formula can supply the nutrients necessary to infant growth, used as a substitute for breast milk. However, according to the third national oral health epidemiological survey show that the caries rate of children under the 5 age was 66.00% in 2005, if the main reason is caused by the bottle feeding.We added erythritol in the formula to satisfy children’s nutritional requirements for sugar intake and reduce the occurrence of caries. We chosen Lactobacillus acidophilus(La) as the research object, further explore the feasible and mechanism of erythritol for caries disease prevention.Methods:1. La was anaerobically cultured(80%N2, 10%CO2, 10%H2) at 37℃ in 5 Erythritol- Formula Milk Powder mixture groups(experimental groups) and 1 with formula milk powder group(control group). And 5 experimental groups respectively in 1%,2%,4%,6%,8% concentration of erythritol. The lactic acid was extracted for HPLC to analyze at 4、6、8、12、16、20、24h,and recorded the peak-area and calculate the concentration(g/L) of lactic acid. At the same time determined the p H of every groups.2. After 18 h cultured in every groups, CFU of La was determined by dilution method of plate counting.3. Dentin cakes have been put into every groups’ liquor with La. After 18 h cultured, dentin cakes were scanned by CLSM and ESEM to observe the situation of adhering by La.Results:1. The concentration of lactic acid is difference between time and time is statistically significant(P﹤0.05),and is not completely equivalent in each groups(F=187.448,P﹤0.05).Concentration of lactic acid of each experimental groups is lower than control group(P﹤0.05); Pairwise comparison(time is without consideration), the difference in 4% E-M, 6% E-M, 8% E-M is not statistically significant, and difference between each of them and others is not statistically significant too(P﹥0.05).2. The CFU of all experimental groups are less than control group, and the difference is statistically significant(P﹤0.05). The difference in 4% E-M, 6% E-M, 8% E-M is not statistically significant, and difference between each of them and others is not statistically significant too(P﹥0.05).3. The green area of MRS, 6%E-M, 8%E-M groups could be observed by CLSM was less than of M, 1%E-M, 2%E-M groups. Comparing corresponding lamina’s live percentage of all groups by statistical, 6% E-M and 8% E-M’s are significant less than M, the difference is statistical significance(P﹤0.05);The EPS and bacterial population of 2%E-M,4%E-M, 6%E-M, 8%E-M is less than of M, 1%E-M by ESEM observing, and the structure of dentin is more clear.Conclusions:Erythritol can cause the inhibition of La’s growing and metabolizing lactic acid in formula milk powder, the effect of erythritol with high concentration(4%,6%,8%)is better than low concentration(1%,2%),and the 6%,8% concentration of erythritol also can dramatically inhibit the La adhere to dentin cakes in formula milk powder.
