In this paper, the different aged pit mud, which were collected from two different regions—Forgood distillery(include aged of 5 years and 30 yearsor so) and Luzhou Laojiaodistillery(include aged of 40 years and 100 yearsor so), were used as the tested subject. The main research contents include: studied on the microbial relationship of these pit mud; discussed its related characters of physical and chemical index and microbial community; analyzed the correlation between its microbial community and physical and chemical index. The results were showed as follows:1. The changes ofphysical and chemical indicators and mineral elements content in pit mud of different pit ages and different regions.Pit mud of 100 years pits agehad significant difference in physical and chemical indicators or mineral elements(P<0.05); The sample of different regions and the same pits age had non-significant difference in physical and chemical indicators(P>0.05), and significant difference in mineral elements(P<0.05).2. Bacterialcommunity structure in pit mud of different regions and different age.(1) In pit mud of Forgood distillery, the test obtained 1136 OTUs, 8 kinds of phylums. The main bacterial phylums were Formicates and Bccteroidetes. The dominant bacterial genus were Sporobacter、Sedimentibacter and Syntrophomonas;(2) In pit mudspf Luzhou Laojiao, the test obtained 998 OTUs, 5 kinds of phylums, the main bacterial phylums were Formicates 、 Bccteroidetes and Chloroflexi. The dominant bacterial genus were the unknown bacterial genus of Sporobacter 、Syntrophomonas、Sedimentibacter and Ruminococcaceae;(3) There were differences in bacterial community structure of the different age pit mud, and the main phylum and genus were different. There are large differences in bacterial community structure in the pit mud of different regions.3. Archaeal community structure in pit mud of different regions and different age.(1)Pit muds ofForgood distillery had 78 OTUs, pit muds ofLuzhou Laojiao had 135 OTUs, the main archaea were Methanobrevibacter 、 MethanocμLleus 、Methanobacterium、Methanosarina and Thermoplasmatales;(2) There were small differences in archaeal community structure in the pit mudof different age, and the main genus were slight different. There are large differences in that of different regions.4. The differences of microbial communities in different age pit mudand its correlation analysisMicrobial community structure of pits had space&time differences. In the same region, the similarity of bacterial and archaeal community structure of the same age pits were higher than that of different age pits. In the same age pits, the similarity of bacterial and archaeal community structure of the same regions were higher than that of different regions’ pits. Physical and chemical indicators were extremely significant correlation with pit ages(P<0.01). Some physical and chemical indicators were extremely significant correlation with bacterial or archaeal diversity(P<0.01).PCA analysis showed that water content was the largest influence factor on microbial community.
