The oligodeoxynucleotides,with the sequence of 5'-CCT CAT GCG GTA AAT TTC GG-3',was synthesized,purified and identified in this research. These Antisense oligodeoxynucleotides(ODNs) can be complementary with a fragment in receptor gene of Insulin like Growth Factor-1 (IGF-1). The ODN chains can specifically hybridized to the messenger RNA of receptor gene of IGF-1,thus,it can inhibit the expression of receptor gene of Insulin like Growth Factor-1. It was reported that the expression of the gene of IGF-1 is in a high level in many kinds of human tumors and cancers. Therefore,the ODNs can inhibit the cancer cells from growing.The chemical procedures of the synthesis cycle composed of four steps,which are deblocking step,activating and coupling step,capping step,oxidation step. After 20 cycles,the final ODN chains was obtained,and then it was purified and identified by methods of 1-butanol,OPC,PAGE,HPLC and others. |
