The research of the pharmacodynamics shows that the total flavone of fructus aurantii imoaturus have the bidirectional adjusting effect to the intestinal smooth muscles. It has low toxicity, large safty range and very distinctly officinal value.This thesis includes two parts:The first part introduces the herbal medicine origin, chemical components, pharmacological effects and clinical applications, and summarizes the differentiation, content determination, physical-chemical characters and the extractive and separate methods of main flavone of fructus aurantii immaturus.The second part can be divided into four aspects:1. Determination of diffirent fructus aurantii immaturus by uv spectrophotometry and HPLC, and the finally choice trueborn fructus aurantii immaturus offered by ZhangShu, JiangXi, that is used in experiment and manufacture.2. Confirmation of the best extractive technics of the flavone of fructus aurantii immaturus through the four factors (concentration of ethanol,time of extraction, dosage of extractive solvent, times of extraction) three levels orthogonal test, according to the charactors of the main flavone of fructus aurantii immaturus. The best extractive technics is that adding 6 times 60% ethanol to the medicinal materials, steeping it for 1 hour;refluxing 2 times, 2 hours per time;then washingit with 10 times waters 2 times, 1 time per day. Consequently, the total flavone content of fructus aurantii immaturus reaches 70%.3. Confirmation of the extractive method of neohesperidin from fructus aurantii immaturus. (the purity is more than 99%,) and the structure through the method of UV, IR, MS,1HNMR.4. Determination of total flavone of the fructus aurantii immaturus. hesperidin and neohesperidin through UV spectrophotometry and HPLC chromatogram.
