Acarbose, as a kind of new α-glucosidase inhibitor has been used in non-insulin-dependent diabetes mellitus therapy. It competitively combined with glucosidase resulting in a inhibition effect on glucosidase , and resulting in a reduced sugar concentration in blood. Because of its complicated chemical structure, industrial production of Acarbose at present is achieved by fermentation of microbe.In this paper, a high efficiency method was established by integrating protoplasts mutation techniques with solid fermentation screening techniques and the OD detecting acarbose methods replacing HPLC method. Applying this method in the procedure of strain-improving of acarbose-producing strain, wonderful efficiency of strain-improving was acquired - the titre of acarbose rearched 3,700μg/mL, 2.2 times as much as that of original strain.First, the condition of preparation and regeneration of protoplasts of Actinoplanes sp. was investigated. After improving the conditions of preparation and regeneration of protoplasts, the concentration of protoplasts reached about 8.0×107/mL, and about 8% regeneration rate of protoplasts was acquired , so the necessary condition of protoplasts mutation was met.Then, the condition of solid fermentation was investigated, a suitable condition for solid fermentation was set up, and a high efficiency method of detecting acarbose produced by solid fermentation was established. By combining these new techniques, a high efficiency strain-improving procedure was set up. This new strain-improving procedure increated the speed of screening greatly and decreased the screening cost obviously.Further more, the suitable mutagen and suitable dosage of mutagen for protoplasts mutagenesis were investigated, and a rational screening model - NH4+ tolerance model was set up.Adopting this new strain-improving procedure, a new mutant AC-N1 tolerating NH4+...
