Zeaxanthin can also decrease the risk of cataract, age-related macular degeneration(ARMD),and cardiovascular disease. In this work,systematic studies were carried out on the isolation, purification, quantification, antioxidant activity, free radical scavenging activity of zeaxanthin from CGM.The optimum conditions of purification by thin-layer chromatography(TLC) was obtained using silica G plate as the support and a solvent mixture of petroleum ether/acetone as the developing solvent. Zeaxanthin was qualitatively identified by various physico-chemical properties such as Rf value from TLC,the color change on reaction with Ag+,UV-visible absorption spectra,the retention time from HPLC,and the molecular weight as measured by MS.The apart component was identified as zeaxanthin.The technology of zeaxanthin preparation was obtained by CGM followed by 95% ethanol extraction and TLC or column chromatography.The parameters of zeaxanthin extraction was optimized .The reacting equation was Y=50.4333+1.075*x1+2.2125*x2+0.9375*x3 -2.0542*x1*x1-0.25*x1*x2-1.0792*x2*x2+1.2*x1*x3-0.575*x2*x3-2.0792*x3*x3Supercritical CO2 fluid extraction is a kind of new extracting technology. Based on the fundamental principle of the technology,extracting pressure,extracting temperature,extracting time were studied.The reacting equation was Y=67.3+2.8*x1+4.45*x2+2.45*x3 -2.925*x1*x1-0.6*x1*x2-1.425*x2*x2-0.45*x1*x3+0.5*x2*x3-3.725*x3*x3The antioxidant properties was investigated by ferric thiocyanate method.Zeaxanthin exhibited high antioxidant activities in the linoleic acid auto-oxidation process.The superoxide anion and hydroxyl radical scavenging capacities of zeaxanthin was investigated by chemiluminescence.
