916 is one kind of sulfated amino polysaccharide which is derived from marine chitin and by ways of multiple molecular modifications.916 shows good antiatherosclerotic activity. This essay using capillary electrophoresis method to investigate the interaction between 916 and Human Serum Albumin, for the purpose of providing a simple, easy implement and feasible approaches for drug-protein binding study.In lab proc, we choose an optimization electrophoresis condition as:pressure injection for 15 seconds, running voltage 15KV, temperature at 25℃,detection at 214nm, concentration of running buffer is 20mmol/L and pH 7.4.Set 916 as buffer addition and set HSA as sample, detect the migration time of HSA, then calculate the inding constant according to Scatchard formula, Ka=2.1×104.We optimized injection time and separation voltage, investigated the precision of peak height and migration and chose an optimization electrophoresis condition as: 67mmol/L phosphate buffer pH 7.4, pressure injection for 25 seconds, running voltage 20KV,temperature at 25℃and detection at 214nm. The result shows that 916 has little affection on statins's binding to HSA, concentration of free drug changes in the range of±6μmol/L, it is lower than 10% of free drugs, there is no substitution between 916 and statins.
