OBJECTIVE:By spectrophotometric method and Maxwell turbidimetry method etc. giving a test on concentration of bacteria.Understanding of the characteristics of E. coli at different growth periods, analysing an effect of different ion concentration on Escherichia coli growth situation. the formation being that the microelement is used for Escherichia coli extracorporeal culture and the formation of biofilm provides reference numerical value. To understand the different iron and zinc ion concentration on the antibacterial activity of azithromycin, Taking silica gel as carrier and building extracorporeal Escherichia coli biofilm model, Understanding of biofilm formation of Escherichia coli, By bacterial counts, silver staining, etc.studing certification standards of biofilm formation,and the effect of Azithromycin in combinatioan with berberine.METHODS:1 With time as the abscissa, With OD value of the vertical axis building coordinate system, respectively measuring OD value of bacterium liquid in different Escherichia coli growth period, drawing out the growth curve of Escherichia coli. Using culture medium to compound the culture medium becoming the zinc, iron ion containing different concentration, By spectrophotometric method after inoculating with, make use of OD value reaction bacterium concentration, building up time-OD coordinate system to analyse an effect of ion on Escherichia coli growth value through the curve toward.2 By disk diffusion method of susceptibility test to compare the different iron ion and zinc ion concentration of azithromycin on the diameter of the impact of E. coli bacteria;3 taking silica gel as carrier, building the Escherichia coli biofilm model.,Comparison with the bacterial count of azithromycin in combination with berberine and individual drug efficacy.RESULTS:1 Escherichia coli growth curve generally consistent with the microorganism growth characteristic.and displays diauxic growth curve characteristic.Iron ion concentration were 11.17,27,32μmol/l respectively, View from the growth curve, by adding iron ions in each group of E. coli bacteria of OD was significantly large compared with the control group. Concentration in the first three groups, with the increasing concentration of OD values increasing, but to 32μmol/L concentration, OD values than 27umol/L concentration of OD values to be small. In this study, comparisons between different concentrations of E. coli in 27μmol/L of iron ion concentration of the OD value of the maximum. Zinc ion concentration were 6,12,18,52μmol/l respectively.Compared with the control group, the concentration of zinc ion group ODvalues are lower than the control group, in general, with increasing concentration, ODvalues increased, but the zinc ions in 12-18umol/L concentration range.OD value is almost no significant change in value is relatively stable.2 In culture medium which contain iron concentrations 11,17,27,32μmol/l inhibition zone diameter further evident that iron ions can promote the growth of E. coli. In the same,it also can evident zinc ions can inhibit the growth of E. coli. but the concentration of ions on the antibacterial activity of azithromycin had no effect.3 Making use of silver dyeing method appraisal, building thecherichia coli biofilm model successfully.0.15,0.3,0.6mg/l of azithromycin and 0.2,0.4,0.8mg/l of berberine, single drug and after each other combination therapy compared bacterial counts after analysis and comparison, when the concentration of berberine 0.8mg/L apparent only synergistic inhibitory effect, and the synergistic effect of azithromycin has nothing to do with concentration.CONCLUSION:Iron ions on the growth of E. coli has some role in promoting, but a great relationship with its concentration. Low concentration of almost no promotion, with the iron ion concentration started to increase its role, but its role to reach a certain concentration, not only will not strengthen but weaken. Normal serum zinc concentration in the 11-23umol/L range inhibit the growth of E. coli, the lack of zinc in the body of the case, the more susceptible to E. coli. Zinc ions in 12-18μmol/L concentration range of roles did not change significantly more stable. But with the zinc ion concentration in the increase to twice the highest concentration of the body a long time, inhibition is rather significant. Different concentrations of iron, zinc ions on the antibacterial activity of azithromycin had no effect. When berberine concentration of 0.8mg/L, berberine azithromycin combined with the biological membrane of E. coli was synergistic inhibitory effect.
