Objective:To identify Taenia Cestodes in 6 regions from 3 counties of Dali City, Yunnan province using morphological and molecular methods. Further to clarified the existence of Taenia Cestodes in the region, to specific categories of Taenia asiatica provide a reference orientation; preventing and treating taeniasis and cysticercosis in Yunnan province.Methods:(1) Morphological Identification:Cutting scolex, mature proglottid and gravid proglottid of taenia cstodes from 6 regions from 3 counties of Dali City, Yunnan province and two control groups from T. solium and the T. saginata, dyeing respectively by Malachite Green Staining and Hydrochloric Acid Staining Red Card, conducted a preliminary identification of Morphology. (2) Molecular biological Identification: Mitochondrial DNA were extracted from the proglottis of adult Taenia cestode strains in 6 regions from 3 counties of Dali City,Yunnan province,partial segments of mitochondrial DNA-cytochrome B(mtDNA-Cytb) sequences were amplified by PCR and then sequenced. Homology matrix, Genetic distances and Phylogenetic tree of species strains was constructed with other known mtDNA-Cytb gene sequences of T. solium T. saginata and Taenia asiatica, in GenBank using DNAMAN (Version 5.2.2) software.Results:(1) By morphological identification, the scolex was rectangular in shape,and had 4 suckers. Neither hooklets nor rostellums were observed. From an mature proglottids perspective, it has two lobe ovaries. Uterine subbranches of gravid proglottids was approximately symmetry on each side, with 13~20 main branches unilaterally. It is similar to Taenia asiatica. (2) Sequence analysis results suggested that mtDNA-Cytb gene sequences of Taenia Cestodes from Eryuan and Suanglang,Wase of Dali and Taenia asiatica gene sequence were fundamentally identical, and the homology reached 99% and 100%. The Heqing of Dali homology with T.solium was 99%. In the phylogenetic tree, EY2,WS and EY1,SL,DY sample clustered into one strain and YZ, are the Taenia asiatica; D Land ND monophyletic group, a traditional T. saginata; HQ1-3 and ZD monopnyletic group, belongs to T. solium; The Taenia asiatica and T. saginata clades were clustered into one clade, then it was grouped to the T. solium.Conclusion:(1) The Taenia cestode samples Eryuan and Suanglang,Wase of Dali in Yunnan province belong to Taenia Cestodes in Taenia asiatica; The Taenia cestode samples Heqing of Dali belongs to T. solium. These data demonstrate that Taenia asiatica is sympatrically distributed with T. solium in Dali. (2) Sequences analyse of mtDNA-Cytb is effective in identifcation of elucidating phylogenetic relationships among species.
